颅咽管瘤中RARα及PPARβ/δ表达与RA作用的相关性

摘要 目的检测颅咽管瘤中RARα及PPARβ/δ的表达,分析其表达量与RA作用效果的相关性,探讨RA靶向治疗颅咽管瘤的分子机制.方法 RT-PCR法检测31例体外培养的颅咽管瘤细胞中PPARβ/δ、RARα的表达情况,MTT法测定RA对PPARβ/δ、RARα不同表达的颅咽管瘤细胞的抑制率,分析其表达情况与RA作用是否存在相关性.结果 RT-PCR结果显示,不同颅咽管瘤细胞中PPARβ/δ、RARα表达存在差别.31例原代培养的颅咽管瘤细胞按其核受体表达不同分为PPARβ/δ>RARα、RARα>>PPARβ/δ、RARα>PPARβ/δ3组;MTT结果显示在相同RA药物作用下RARα>PPARβ/δ组的抑制率明显高于RARα>PPARβ/δ组、PPARβ/δ>RARα和对照组,差异有统计学意义(P<0.01).结论 PPARβ/δ、RARα的表达可作为评估RA治疗颅咽管瘤效果的有效指标.PPARβ/δ表达低的颅咽管瘤细胞对RA更敏感;靶向升高RARα或者靶向降低PPARβ/δ的表达都有益于颅咽管瘤的治疗. Objective To detect the expression of Retinoic acid receptor alpha (RAR α ) and peroxisome proliferator-activated receptor β/δ (PPAR β/δ) in craniopharyngioma cells, analyze the correlation between the expression and effect of retinoic acid (RA) , and investigate the molecular mechanism of RA targeted therapy for craniopharyngioma. Methods The expression of RAR α and PPAR β/δ in 31 cases of craniopharyngioma cells in vitro was quantified by reverse transcription-PCR. The inhibition rates of RA in craniopharyngioma cells with different expression levels of RAR α and PPAR β/δ were detected by using MTT assay. And the correlation between expression and effect was analyzed. Results The RT-PCR results showed that the expression of PPAR β/δand RAR α mRNA in craniopharyngioma cells was different. 31 cases of craniopharyngioma cells in primary culture were divided into three groups according the expression differences of nuclear receptor: PPAR β/δ 〉 RARα, RARet 〉 PPAR β/δ and RARα〉〉 PPARβ/δ. MTT results showed that the inhibition rate of RAR α 〉〉 PPAR β / group was significantly higher than the other groups under the same condition, with statistical significance (P 〈 0.01) . Conclusion The expression of PPAR β/δand RARα can be used as effective index to evaluate the effect of RA in craniopharyngioma. The craniopharyngioma with low-expression PPAR β/δ was more sensitive to RA.Targeting higher RAR αor targeting lower PPARβ/δ is beneficial to the treatment of craniopharyngioma.
