摘要
目的探讨p53和MDM2在胃腺癌组织中的表达及临床意义。方法采用免疫组织化学染色方法检测94例胃腺癌组织中p53和MDM2的表达水平,分析两者表达的相关性。结果 p53(突变型)的检出率为36%(34/94),在这些可检出突变型p53蛋白表达的胃腺癌原发灶组织中并非所有癌细胞均为p53阳性表达,其阳性率差异较大,从10%99%,均数约30%;在62.8%(59/94)的胃腺癌原发灶组织中可检测到MDM2蛋白表达,定位于胞质和胞核,呈异质性表达;MDM2蛋白表达水平在p53野生型胃腺癌组织(免疫组织化学染色阴性)中高于p53突变型胃腺癌组织(免疫组织化学染色阳性)。结论在胃腺癌组织中,p53和MDM2呈异质性表达;在部分胃腺癌细胞中,高水平MDM2蛋白表达是限制野生型p53蛋白积聚活化的主要原因之一。
Objective To investigate the expression and clinical significance of p53 and MDM2 in gastric adenocarcinoma. Methods The protein expressions of p53 and MDM2 in 94 gastric adenuearcinoma tissues were detected by immunohistoehemical staining, and the relationship between the two proteins was analyzed. Results Mutant - p53 protein was detected in 36% (34/94) of primary gastric adenoearcinoma, and the percentage of p53 - positive cells was highly variable ranging from 10% to 99% ( median, 30% ). MDM2 protein was located in the cytoplasm and nucleus of gastric adcnocarcinoma cells, and the positive rate of MDM2 expression was 62.8% (59/ 94 ). The expression level of MDM2 in wildtype - p53 gastric adenocarcinoma tissues ( negative immunohistoehemical staining) was higher than that in mutant - p53 tissues ( positive immunohistoehemical staining). Conclusion Variable expressions of p53 and MDM2 were detected in gastric adenocarcinoma tissues, and high expression level of MDM2 protein in part of gastric adenocareinoma cells may be one of the main reasons that contributes to the limitation of wildtype - p53 accumulation and activation.
