四妙勇安汤对动脉粥样硬化大鼠氧化损伤的保护作用 被引量:14

摘要 目的:探讨四妙勇安汤对AS大鼠脂质代谢的影响,为临床用药提供依据;方法:将SD雄性大白鼠40只随机分为假手术组、空白模型组、四妙勇安汤组、普罗布考组,每组10只。制备AS模型,四妙勇安汤按15.22g/kg给药,普罗布考组按0.16g普罗布考/kg体重给药,假手术组、空白模型组给予同体积的生理盐水。于实验16周末禁食16h后,麻醉动物,腹主动脉取血5ml留置2管,分别分离血清,观察血清中谷胱甘肽过氧化物酶(GSH)、超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)、抗总氧化物能力(AOC)、黄嘌呤氧化酶(XOD)的变化;腹主动脉取血后,分离颈总动脉,在主动脉弓起始段取0.5cm主动脉标本做病理切片,观察动脉管壁形态学变化。结果:四妙勇安汤组可以显著降低AS大鼠血清TC和LDL-C水平(P<0.01);可以有效地保护血管内皮,防止AS的形成;升高AS大鼠血清GSH、NOS、T-AOC和SOD水平(P<0.05,P<0.01),降低XOD水平(P<0.01);结论:四妙勇安汤可以舒张血管,清除氧自由基,保护血管内皮,防止AS的发生,其机制与升高GSH、NOS、T-AOC和SOD水平、降低XOD水平有关。 Objective:To explore the mechanism of Simiao soup on lipid metabolism of atheroselerosis (AS) rats.Methods:Forty male SD rats were randomly divided into sham operation group,untreated group,Simiao soup group and probucol group (n=10).Preparation of the AS model,each group modeling 3rd day administra- tion Simiao soup 15.22g/kg were given in Simiao group,probucol 0.16g/kg were given in the probucol group. The rats of sham-operated group,blank model group were given the same volume of saline.After 16h fasting at the end of 16 weeks,anesthetized animals,abdominal aortic blood 5ml indwelling 2 tubes,respectively.The serum was separated and observed glutathione peroxidase(GSH),superoxide dismutase(SOD),nitric oxide syn- thase(NOS),total antioxidative capacity(AOC)and xanthine oxidase(XOD).The initial segment(0.5cm)of carotid artery and aortic arch were harvasted and underwent pathologic examination.Results:Simiao soup group can significantly reduce serum TC and LDL-C level (P〈0.01)of AS rats ;can effectively protect the vascular en- dothelium;elevate serum GSH,NOS,T-AOC and SOD levels(P〈0.05,P〈0.01),reduce the the XOD level of(P〈 0.01)AS rats.Conelusion:Simiao soup can relax blood vessels,scavenge oxygen free radicals,protect the vascu- lar endothelium,prevent the occurrence of AS.The mechanism associates with elevated GSH,NOS,T-AOC and SOD levels,reduce XOD level.
