摘要
【目的】观察痹痛康丸对胶原性关节炎(Collagen-induced Arthritis,CIA)大鼠踝关节滑膜白细胞介素1β(IL-1β)的影响,探讨其治疗作用机制。【方法】建立II型胶原加完全弗氏佐剂诱导的大鼠CIA模型,70只Wistar大鼠随机分为7组,分别为痹痛康丸低、中、高剂量组[0.3、3、30 g.(kg.d)-1],尪痹冲剂组3.g(kg.d)-1,甲氨蝶呤片组1.2 mg.(kg.w)-1,以上各组药物溶解于2 ml生理盐水中,1次/d,连续灌胃给药4周。空白对照组及模型组灌服生理盐水2 ml/只。8周后处死大鼠,观察右踝关节滑膜病理改变情况及关节滑膜中IL-1β蛋白表达量的变化。【结果】各用药组除痹痛康丸低剂量组外滑膜组织的病理改变均有不同程度的减轻,IL-1β蛋白的表达量较模型组降低(P<0.05)。【结论】痹痛康丸能有效降低CIA大鼠滑膜炎程度,防止骨破坏或关节重构,有免疫调节作用。
【Objective】To investigate the impacts of Bitongkang pill on IL-1β in collagen-induced arthritis(CIA) rats ankle joint,and to explore its therapeutic mechanism.【Methods】Model of type II collagen induced arthritis established in Wistar rats.Rate were randomly divided into seven groups: they were Bitongkang pill low,medium and high dose group [0.3,3,30 g.(kg.d)-1],Wangbi diluent group 3 g.(kg.d)-1,Methotrexate piece group 1.2 mg.(kg.w)-1,the drugs of above groups were dissolved in 2 ml of saline,administrated intragastrically once per day for 4 weeks.Blank control group and model group,were fed with 2 ml saline/rat.After eight weeks,the rats were sacrificed and the pathological changes of the right ankle joint synovial and synovial the amount of expressed IL-1β were observed【.Results】The pathological changes of the synovial tissue were lightened in varying degree in all the treated group except the Bitongkang pill low dose group.The amount of IL-1β protein expression decreased in all the trested group(P 0.05)【.Conclusion】 Bitongkang pill may effectively reduce the degree of CIA rats synovitis,prevent bone destruction or joint reconstruction,and have immunomodulatory effects.
