摘要
以培养25d的狗头枣试管苗叶片为材料,研究了不同酶液浓度,不同甘露醇浓度对狗头枣组培苗叶片原生质体分离的影响。结果表明适合狗头枣组培苗叶片原生质体分离的适宜酶液配比为1.0 g/L纤维素酶、0.4 g/L果胶酶,0.6 mol/L甘露醇,黑暗条件下,酶解8 h,可获得大量有活力的原生质体。其叶片原生质体的产量为2.12×106个/g,活原生质体获得率为80.88%。
Study on the influence of different enzyme concentrations and different concentrations ot man- nitol on protoplast isolation in Chinese date Goutouzao leaf tissue culture seedling by 25 d leaves. The results show that suitable for tissue culture seedling of Chinese date Goutouzao leaf protoplast isolation was 1.0 g/L cellulase,0.4 g/L pectinase and 0.6 mol/L mannitol, dark conditions, enzymatic hydrolysis of 8h, can obtain a great amount of viable protoplasts. The protoplast yield was 2.12 x 10^6 pre/g, living protoplast obtained rate 80.88%.
