摘要
目的探讨趋化因子CCL20及CCL22在狼疮鼠浆膜腔液中的表达及意义。方法以9只B6.MRL-Faslpr/J狼疮小鼠为观察组,6只同系C57BL/6J小鼠为对照组。取2组小鼠胸膜及腹膜腔液,用ELISA方法检测其中CCL20及CCL22的表达水平,并与小鼠血清中ANA、dsDNA抗体的含量以及小鼠24 h尿蛋白含量进行关联性分析。结果 CCL20在狼疮鼠浆膜腔液中高表达,与对照组相比两者差异有统计学意义(P<0.05),而CCL22与对照组相比两者差异无统计学意义(P>0.05);狼疮小鼠浆膜腔液CCL20含量与血清ds-DNA和24 h尿蛋白量呈正相关(P<0.05),与血清中ANA无明显相关性(P>0.05);而狼疮小鼠浆膜腔液CCL22含量与血清ds-DNA、ANA及24 h尿蛋白量均无明显相关性(P>0.05)。结论趋化因子CCL20可能参与狼疮鼠浆膜腔病变的发生,与狼疮鼠的病情活动、肾损害有一定相关性。
To evaluate the expression and significance of CCL22 and CCL20 in the serous membrane effusion of lupus mouse, nine B6.MRL-Fas^lpr/J lupus mice were selected as experimental group and six C57BL/6J mice as healthy controls. CCL20 and CCL22 levels in serous membrane effusion were assessed by enzyme linked immunosorbent assay (ELISA) for analyze their correlation with serous ANA, serous ds-DNA antibody, and 24 h urine protein in lupus mice. ELISA showed CCL20 level of serous membrane effusion was significantly increased in lupus mouse compared with that in healthy controls (P〈 0.05), but CCL22 level did not show difference (P〉 0.05). The CCL20 of serous membrane effusion was positively correlated with serum ds-DNA antibody and the 24 h urine protein (P〈0.050), but not correlated with serum ANA (P〉 0.05). The CCL22 of serous membrane effusion was not correlated with serum ANA, serum ds-DNA, or 24 h urine protein. In conclusion, CCL20 may be involved in the oromeningitis of lupus mouse and may be correlated with the pathogenesis and disease activity of lupus mouse.
