摘要
目的:探讨蛋白酶体抑制剂硼替佐米对人绒毛膜癌细胞株JEG-3细胞增殖、凋亡及核因子κB(NF-κB)表达的影响。方法:设空白对照组,以不同浓度(0.1~200nmol/L)硼替佐米作用于JEG-3细胞,采用CCK-8法检测细胞增殖抑制率,以荧光显微镜检、流式细胞仪检测细胞凋亡,免疫荧光法检测细胞NF-κB的表达。结果:不同浓度的硼替佐米作用于人绒毛膜癌JEG-3细胞48小时,细胞增殖抑制率随着药物浓度增加逐渐增强(P<0.05)。以10nmol/L硼替佐米作用于JEG-3细胞24小时、48小时、72小时,随着作用时间延长细胞增殖抑制率逐渐增强(P<0.01)。与对照组比较,10nmol/L硼替佐米作用于JEG-3细胞48小时,荧光显微镜下见细胞核固缩、核碎裂及凋亡小体,AnnexinV-FITC/PI双标法显示细胞凋亡率增高(P<0.01)。免疫荧光结果显示硼替佐米组凋亡细胞中NF-κB表达增强。结论:在一定浓度、时间内,硼替佐米呈浓度、时间依赖性抑制人绒毛膜癌JEG-3细胞的增殖,诱导其凋亡,且细胞凋亡可能与NF-κB表达增强有关。
Objective :To investigate the effect of proteasome inhibitor bortezomib on proliferation,apopto- sis and the expression of NF-KB of human choriocarcinoma JEG-3 cells. Methods:JEG-3 cells were treated with bortezomib at different concentrations(0. 1 ~200 nmol/L). Cell proliferation was analyzed by CCK-8 assay,cell apoptosis by flow cytometry and fluorescence microscope, NF-KB protein expression by immunofluorescence, blank control was set. Results:JEG-3 cells were treated with bortezomib at different concentrations for 48 h respectively,the cell proliferation inhibition rate gradually increased with the increase of drug concentration( P 〈 0.05). JEG-3 cells was treated with bortezomib(10 nmol/L) for 24 h,48h and 72 h,cell prolifera tion inhibition rate gradually increased with the prolonging of drug action time (P 〈0. 01 ). Compared to the control group,JEG-3 cells treated with 10 nmol/L bortezomib for 48 h showed nuclear condensation, nuclear fragmentation,apoptotic body formation and increase of the apoptotic cells rate( P 〈0. 01 ). The expression of N F-KB in apoptotic cells after bortezomib treatment was significantly increased. Conclusions: Bortezomib can inhibit the proliferation of JEG-3 cells,and induce apoptosis concentration and time dependently in a certain scope of concentration and reacting time,which maybe related to the increasing the NF-KB expression.
