摘要
目的:探讨温阳活血方对慢性肾衰竭(chronic renal failure,CRF)大鼠肾脏中转化生长因子β1(transforming growth factor-β1,TGF-β1)表达的影响。方法:选用120只SD雄性大鼠,通过5/6肾切除建立慢性肾衰竭大鼠模型。随机分为5组:(1)假手术组:予生理盐水10ml·kg-1·d-1灌胃;(2)模型组:予生理盐水10ml·kg-1·d-1灌胃;(3)中药组:予温阳活血方30g·kg-1·d-1灌胃;(4)西药组:科素亚33.3ml·kg-1·d-1灌胃;(5)中西药联合组:予温阳活血方+科素亚灌胃。分别在给药后4、8、12周,每组随机选取8只大鼠处死,检测肾功能指标和24h尿蛋白,用免疫组化法检测肾脏TGF-β1表达水平。Western印迹检测给药12周后TGF-β1蛋白在肾组织中的表达。结果:与假手术组比较,模型组大鼠血BUN、Scr、24h尿蛋白定量明显均明显升高(P<0.01,P<0.05);三个治疗组血BUN、Scr、24h尿蛋白定量明显低于模型组(P<0.01)。免疫组化显示,治疗组均能显著减少TGF-β1在肾脏中的表达(P<0.01)。结论:温阳活血方可能是通过降低慢性肾衰竭大鼠的Scr、BUN和尿蛋白水平,抑制肾脏TGF-β1表达,从而延缓肾衰竭进展。
Objective:To study effect of Wenyanghuoxuefang(WYHXF) on transforming growth factor - β1 (TGF - β1 ) expression in kidney tissue of rats with chronic renal failure(CRF). Methods:After one week of 5/6 Nephrectomy , male SD rats were randomly divided into five groups. Shamoparation group were treated with normal saline( 10 ml·kg^-1·d^-1). Model group were treated with normal saline( 10 ml·kg^-1·d^-1). Wenyanghuoxuefang group were treated with WYHXF(30 mg·kg^-1·d^-1 ). Losartan group were treated with Losartan Potassium(33.3 mg·kg^-1·d^-1). Wenyanghuoxuefang combined Losartan group were treated with WYHXF (30 mg·kg^-1·d^-1 ) and Losartan Potassium (33.3 mg·kg^-1·d^-1 ). After the treatment,eight rats in each group were killed in 4 th, 8 th and 12 th weeks. Scr, BUN and 24h - urine protein were measured the level of TGF - β1 expression were determined by immunohistochemical method in the meantime. Western blot was used to test TGF - β1 protein in 12 th weeks. Results: Compared with Sham - oparation group, the level of 24 h - urine protein, Scr and BUN were significantly increased in model group (P〈0. 01 ,P 〈0.05 ). Compared with model group, the level of 24 -urine protein, Scr and BUN were significantly decreased in three treatment group (P 〈 0. 01, P 〈 0.05 ). Immunohistoche - mical technique and computer assisted image analysis showed that the expresstion of TGF - β1 in rats decreased in three treatment group (P 〈 0.01 ). Conclusion: WYHXF could reduce the level of 24 hour urine protein, Scr and BUN and the expression of TGF - β1, and prot - ect renal function and abate the glomerular pathological injury.
