摘要
目的探讨异硫氰酸荧光素-葡聚糖(FITC-Dextran)联合异凝集素(isolectin B4)在评估氧诱导视网膜病变(oxygen-induced retinopathy,OIR)小鼠视网膜新生血管和无灌注区中的作用。方法 10只生后第17天的OIR小鼠分别眶后注射FITC-Dextran,取出视网膜行isolectinB4染色,应用ImagePro-Plus5.1软件测量视网膜新生血管、无灌注区及整个视网膜面积,计算视网膜新生血管和无灌注区占整个视网膜面积的百分比并比较。结果 FITC-Dextran评估的视网膜新生血管占整个视网膜面积的百分比为(0.1009±0.0010)%,均小于FITC-Dextran联合isolectin B4的(0.1046±0.0010)%或isolectin B4的(0.1049±0.0020)%(均为P<0.01)。FITC-Dextran评估的视网膜无灌注区占整个视网膜面积的百分比为(0.2850±0.0010)%,均大于FITC-Dextran联合isolectin B4的(0.2668±0.0010)%或isolectin B4的(0.2676±0.0020)%(均为P<0.01)。FITC-Dextran联合isolectin B4比isolectin B4更易于鉴别视网膜铺片上残留的玻璃体血管,比FITC-Dextran更易于发现未充盈的视网膜小血管。结论 FITC-Dextran联合isolectin B4视网膜血管染色比FITC-Dextran或isolectin B4更能精确评估视网膜新生血管和无灌注区面积。
Objective To investigate the effect of FiTC-dextran combined with isolectin B4 on oxygen-induced retinopathy mice in retinal neovascularization and non perfusion. Methods A total of ten OIR mice at 17th day after being born received an retro-orbital injection FITC-dextran and then their retina was labeled with isolectin B4. The areas of retinal neovascularization,non perfusion,and the total retinal were measured by Image Pro-Plus 5. 1 software. Their percentages were calculated and analyzed. sultu The percentage of the retinal neovascularization in total retinal area evaluated by FITC-Dextran (0. 100 9 ±0.001 0)% was less than those of FITC-dextran combined with isolectin B4 (0. 104 6±0. 001 0)% and isolectin tM(0.10Ll 9 ±0.002 0)% (both P〈0.01). The percentage of retinal non perfusion in total retinal area evaluated by FITC-Dextran (0. 285 0 ±0.001 0)% was larger than those of FITC-dextran combined with isolectin 134 (0.266 8±0. 001 0)% or isolectin tM (0.267 6 ±0.002 0)% (both P〈0.01). It was easier to identify the residual vitreous blood vessels with the method of FITC-dextran combined with lsolectin B4 than that with isolection B4 and it was also easier to find the unfilled small retinal vessels than that with FITC-dextran. neion It is more accurate to e valuate the areas of retinal neovascularization and non perfusion with the method of FITC-dextran combined with isolectin tM than that with FITC-dextran or isolectin B4.
