摘要 目的:综述分析国内外Wnt信号通路和肿瘤干细胞在肿瘤复发转移中的作用的研究现状,为基础与临床研究提供研究方向、思路和资料。方法:应用PubMed、MEDLINE、CNKI以及万方等数据检索系统及数据库,以"Wnt信号通路、肿瘤干细胞、EMT"等为关键词,检索2002-01—2012-03与肿瘤转移相关的中文及西文文献。纳入标准:1)在肿瘤转移过程中的Wnt信号通路以及肿瘤干细胞的基础与临床研究;2)Wnt信号通路、肿瘤干细胞及EMT在肿瘤转移中相互联系的基础与临床研究。根据纳入标准,粗选148篇文献,最后28篇纳入综述分析。结果:肿瘤干细胞(cancer stem cells,CSCs)是指实体肿瘤组织内存在的一群数目极少、但具有无限增殖和自我更新潜能的细胞群体。肿瘤干细胞的存在可能解释了肿瘤的耐药性和不可治愈性,而转移是肿瘤患者死亡的主因。转移的建立取决于肿瘤细胞获得迁移显型的能力以及它们在远处组织重建继发肿瘤的能力。研究表明,EMT(epithelial-mesenchymal transition)在促进肿瘤细胞转移的同时,也使其具有自我更新能力等干细胞样特性。Snail等转录因子、WNT信号通路部分组分在早期肿瘤转移中过表达。结论:研究Wnt信号通路、EMT及肿瘤干细胞在肿瘤转移中的作用机制,将为肿瘤复发和转移的靶向治疗提供新的思路。 Objective : To review recent advances in the role of Wnt signaling pathway and cancer stem cells in tumor recurrence and metastasis. Methods: Chinese and western papers on tumor recurrence and metastasis, published from Jan. 2002 to Mar. 2012, were retrieved in PubMed, MEDLINE, CNKI, and WangFang databases using the keywords Wnt signaling pathway, cancer stem cells and EMT ( epithelial - mesenchymal transition). Criteria of paper adoption : ( 1 ) The foundation and clinical research of Wnt signaling pathway and cancer stem cells in the process of tumor recurrence and metastasis. (2) The foundation and clinical research of the relationship of Wnt signaling pathway, cancer stem cells and EMT in tumor recurrence and metastasis. After that, 148 papers were reviewed, and 28 papers out of them were finally analyzed and reviewed. Results : Cancer stem cells is a group of cells population in the tumor organization with extremely few number, but has the uncontrolled proliferation and self - renewal potential function. The existence of cancer stem cells may explain the resistance of the tumor and incurable characteristics, while the transfer is the main cause of death by cancer patients. The establishment of metastasis depends on the ability of cancer cells to obtain a migratory phenotype and the ability to recreate a secondary tumor in a distant tissue. Studies have shown that EMT can facilitate the invasion and metastasis of cancer cells, and confer the metastatic cancer cells self - renewal ability of stem cells. Recent studies have revealed that several transcription factors such as snail, several components of the WNT signaling pathway were involved in this process and over - expressed in early phase transfer. Conclusion : Here we summarize the recent progress on the roles of Wnt signaling pathway, EMT and cancer stem cells in tumor metastasis, hoping to provide new insights in target therapy of cancer metastasis and recurrence.
