摘要
目的 检测脑胶质瘤组织中端粒酶活性及其亚单位的表达 ,研究端粒酶活性与各亚单位表达的相关性。方法 对 6例正常脑组织和 5 1例胶质瘤组织利用端粒重复序列扩增技术 (TRAP)检测了端粒酶的活性 ,利用PT PCR法对端粒酶不同亚单位的表达情况进行研究。结果 端粒酶活性检测 6例正常脑组织均为阴性 ,5 1例胶质瘤组织中 19例阳性。hTR亚单位在 6例正常脑组织中 5例为阳性 ,在 5 1例胶质瘤标本中 48例为阳性。hTRT亚单位在全部正常脑组织中为阴性 ,在 5 1例胶质瘤中 2 1例呈阳性。TPI亚单位在 6例正常脑组织中均呈阳性 ,在 5 1例胶质瘤中 38例呈阳性。端粒酶阳性与hTRT亚单位表达存在明显相关 ,而与hTR和TP1亚单位表达不相关。结论 部分脑胶质瘤中存在端粒酶的活性 。
Objective To detect telomerase activity and telomerase subunits expression in glioma samples, to study the correlation of telomerase activity and its subunits expression.Methods In 6 samples of normal brain tissue and 51 samples of glioma tissue, TRAP method was used detect telomerase activity, RTPCR method was used to detect the expression of its three subunits. Results Telomerase activity could not be detected in any of the 6 normal brain tissue samples, while 19 glioma tissue samples showed telomerase activity(373%). In the 6 normal brain tissue samples, 5 cases expressed hTR, none of them expressed hTRT and all cases expressed TP1/TLP1. In the 51 glioma samples, hTR expression were found in 48, hTRT expression in 21 and TP1 expression in 38, Correlation study showed that the subunit hTRT had a strong correlation with telomerase activity in glioma samples(P<0005), whereas hTR and TP1/TLP1 expression showed no correlation with telomerase activity.Conclusion Parts of glioma samples show telomerase activity. hTRT subunit may be the ratelimiting subunit in the activation of telomerase.
