Effect of synuclein-gamma on malignant progression in human prostate cancer

摘要 目的观察γ-突触核蛋白(SNCG)在前列腺癌的表达及恶性进展中的作用。方法应用逆转录-聚合酶链反应(RT—PCR)、Westernblot检测SNCG在前列腺细胞株的表达。组织免疫组织化学检测正常前列腺组织、前列腺炎、良性前列腺增生、高级别前列腺上皮内瘤及激素依赖性前列腺癌组织的SNCG表达。结果SNCG在激素依赖性前列腺癌细胞株CWR22、LNCaP的表达明显高于前列腺上皮细胞株RWPE—γ。SNCG在10例正常前列腺组织(1/10)、10例前列腺炎组织(0/10)及10例良性前列腺增生组织(2/10)几乎不表达;在高级别前列腺上皮内瘤(HGPIN)中低表达(3/10);在122例激素依赖性前列腺癌组织有74例阳性表达。SNCG在激素依赖性前列腺癌中的阳性率为60.66%(74/122),其高表达与前列腺癌中高危因素[前列腺特异抗原(PSA)≥10μg/L、Gleason评分≥7、临床分期≥Tb组]及淋巴结转移相关。结论SNCG在激素依赖性前列腺癌中的异常表达,可能参与前列腺癌恶性进展。 Objective To study the expression and significance of synnclein-gamma (SNCG) in human prostate cancer. Methods SNCG expression in the prostate epithelial cell line ( RWPE-I ) and the hormone-dependent prostate cancer cell lines (LNCaP, CWR22) was detected by using reverse tran- scriptase-polymerase chain reaction (RT-PCR) and Western blotting. The immunohistochemistry was per- formed to investigate the expression of SNCG in the normal prostate tissue, prostatitis tissue, benign prosta- tic hyperplasia (BPH) tissue, high-grade prostatic intraepithelial neoplasia, and hormone-dependent pros- tate cancer (HDPC) tissue. Results The expression of SNCG in hormone-dependent prostate cancer cell line was significantly higher than that in prostate epithelial cell line. Almost no SNCG was detectable in be- nign prostate specimens (10 normal prostate tissues, 10 prostatitis tissues, and 10 BPH tissues), SNCG was partially expressed in high-grade prostatic intraepithelial neoplasia (3/10) , and the expression was el- evated to a high level in HDPC tissues (the positive rate 60. 66%, 74/122). Combined with the SNCG stain score and the clinicopa, hological parameters of the corrresponding PCa patients, we found that SNCG protein expression was associated with middle-high-risk PCa characterized by prostate specific antigen (PSA) ≥10μg/L, Gleason score ≥7, and clinical stage≥T2b, and correlated with lymph node metastasis. Conclusion These resuhs suggested that abnormal expression of SNCG may be involved in malignant progression of PCa.
