摘要
目的探讨TIP30在脑胶质瘤中的表达及其临床意义。方法采用免疫组织化学法检测70例脑胶质瘤病人石蜡病理切片中TIP30蛋白表达,并分析其表达与临床病理特征之间的关系;同时,采用RT-PCR方法检测16例脑胶质瘤新鲜组织中TIP30基因的含量。结果 TIP30蛋白在36例(51.4%)脑胶质瘤标本和8例(88.9%)正常脑组织标本中的表达差异有统计学意义(P=0.04)。TIP30的表达与患者的年龄、性别无关,与脑胶质瘤的病理类型、分级和肿瘤大小相关(P<0.05)。单因素分析结果显示,TIP30阳性表达的脑胶质瘤病人总生存时间长于阴性表达组(P=0.003)。RT-PCR结果亦表明,脑胶质瘤中TIP30基因含量低于正常脑组织(P=0.001)。结论 TIP30在脑胶质瘤中表达下调,可能充当抑癌基因的角色,同时也可能是脑胶质瘤病人重要的预后指标之一。
Objective To explore the expression of TIP30 in glioma and its clinical significance. Methods The expressions of TIP30 in paraffin-embedded specimens of 70 glioma patients were detected by immunohistochemistry and its relationship with clinicopathological factors were further investigated. In addition, the relative gene expressions of TIP30 were determined by RT-PCR in 16 glioma tissues. Results There were 36 glioma cases (51.4%) showed positive in TIP30 immunohistoehemical staining, while 8 out of 9 (88.9%) normal brain tissues showed positive. The difference was significant (P=0.04). The TIP30 expression was associated with tumor histology, pathologic grades and tumor sizes (P〈 0.05 ).Survival analysis showed that glioma patients with positive TIP30 expression had a significantly longer overall survival time than those who with negative TIP30 expression (P=0.003). Result of RT-PCR also confirmed that expression of TIP30 gene in glioma tissues was significantly lower than normal brain tissues (P=0.001). Conclusion TIP30 is down-regulated in glioma. It may act as a tumor suppressor gene and possibly be an important biomarker in predicting the prognosis of glioma patients.
