摘要
目的探索骨髓来源细胞(bone marrow-derived cells,BMDCs)能否迁移至舌,并分化为构成舌的各种细胞。方法采用骨髓移植方法构建具有绿色荧光蛋白(green fluorescent protein,GFP)骨髓的C57BL/6嵌合小鼠模型,通过观察胫骨切片中的GFP阳性细胞的分布,判断模型是否构建成功。之后利用嵌合鼠模型,采用免疫组化的方法评价骨髓移植后1、3、6个月时BMDCs在舌内的分化情况。结果骨髓移植后1个月,接受移植的小鼠骨髓内充满了GFP阳性细胞,即建立了嵌合鼠模型。免疫组化结果显示,在嵌合鼠舌内散在分布着大量GFP阳性细胞,包括上皮细胞、朗格汉斯细胞、内皮细胞及结缔组织细胞。结论 BMDCs能够通过循环迁移到舌,在舌内散在分布,并分化为构成舌的多种细胞,参与舌的更新,这为BMDCs应用于舌的再生治疗提供了初步依据。
Objective To investigate whether bone marrow-derived cells (BMDCs)can migrate to tongue and differentiate into the cells of tongue. Methods A green fluorescent protein (GFP) bone marrow -C57BIZ6 chimeric mouse model was firstly built by bone marrow trans- plantation (BMT). Then, the model was assessed by the distribution of GFP positive cells in tibia bone section. At last, immunohistochemistry was performed on tongue sections of 1,3 and 6-month post BMT. Results The bone marrow of 1- month post BMT mice was full of GFP positive cells, conforming BMT success, which meant that the chimeric mouse model had been built. Immunohistochemical analysis indicated a lot of GFP positive cells including epithelial cells, Langerhans cells, endothelial cells and connective tissue ceils were distributed dispersed- ly in the tongues of chimeric mice. Conclusion BMDCs can migrate to tongue, diffe.rentiateinto muhiple cells of tongue and participate in tongue renewal. This could be a preliminary ba- sis for BMDCs application in tongue regenerative therapy.
