单花橐吾愈伤组织诱导及分化培养条件研究 被引量:4

摘要 分别以单花橐吾无菌苗的子叶、真叶、叶柄、根为外植体,进行组织培养的初步研究。结果表明:单花橐吾愈伤组织诱导的最佳外植体是叶柄,最佳诱导培养基配方是MS+6-BA 3.0mg/L+NAA 0.2mg/L+蔗糖35g/L+琼脂8.6g/L,出愈率可达99.5%,诱导效果较好;在MS+6-BA 1.5mg/L+NAA 0.1mg/L+蔗糖30g/L+琼脂8.6g/L中进行增殖培养可诱导出大量不定芽,成苗率可高达98.7%,每块愈伤组织平均可产生7.8个不定芽,且不定芽生长健壮;在1/2MS+NAA 0.5mg/L+IBA 0.2mg/L+蔗糖30g/L+琼脂8.6g/L中进行生根培养,生根率可达99.7%,根白色粗壮,有细绒毛,苗生长健壮。 The tissue culture was carried out by using the cotyledon, euphylla, leafstalk and root of Ligularia jarnesii. The results showed that the best explant for inducting callus was leafstalk, and the op- timum culture medium was MS + 6-BA (3. 0rng/L) + NAA (0. 2rng/L) + sucrose (35g/L) + agaragar (8.6g/L), the callus rate was 99.5%, and the callus grew well; numerous adventitious buds were induced in medium MS+6-BA(1.5rng/L)+NAA(0. lrng/L)+ sucrose(30g/L)+ agaragar(8. 6g/L), stocking percent was 98.7% ,and on the average it could induce 7.8 stuggy adventitious buds per plot of callus. The best rhizogenesis medium was 1/2 MS+NAA(0.5rng/L) +IBA(0.2rag/L) +sucrose(30g/L) + agaragar (8.6g/L), the rooting rate reached 99.7%, the descending axis was white, thick with fine pubescence, and grew well.
