摘要
目的观察延龄草总皂苷对结肠癌细胞HT-29、SW-620的增殖、环氧合酶2(cyclooxygenase 2,COX-2)表达与功能及β-连环素(β-catenin)表达的影响。方法培养结肠癌细胞HT-29与SW-620,加入不同浓度的延龄草总皂苷(0~300.00mg/L),采用四甲基偶氮唑盐(microculture tetrazolium,MTT)比色法,观察延龄草总皂苷对HT-29、SW-620增殖的影响,以蛋白质印迹法(Western blot)检测细胞中COX-2、COX-1和β-catenin蛋白的表达,以酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)方法检测细胞培养液中前列腺素E2(prostaglandin E2,PGE2)的浓度。结果延龄草总皂苷可有效抑制HT-29、SW-620的增殖,其IC50分别为:(9.87±1.15)mg/L,(36.63±1.07)mg/L。延龄草总皂苷可剂量依赖性地降低COX-2、β-catenin的表达和PGE2的浓度。结论延龄草总皂苷可有效抑制结肠癌细胞HT-29、SW-620增殖,可能机制为其降低了COX-2的表达及功能与β-catenin的表达。
Objective To investigate the effects of Trillium saponins on the proliferation of human colon carcinoma cells HT-29 and SW-620,the expression and function of cyclooxygenase 2(COX-2) and the expression of β-catenin.Methods HT-29 and SW-620 cells were administrated with different concentrations of Trillium saponins(0-300.00 mg/L).Cell proliferation was then evaluated by microculture tetrazolium(MTT) assay.COX-2,COX-1 and β-catenin expressions were assessed by Western blot method.Prostaglandin E2(PGE2) concentration was detected by using enzyme-linked immunosorbent assay(ELISA).Results Trillium saponins could significantly inhibit the growth of HT-29 and SW-620,and the IC50 values were(9.87±1.15) mg/L and(36.63±1.07) mg/L respectively.Trillium saponins could reduce COX-2 and β-catenin expressions and PGE2 production in a concentration dependent manner.Conclusion Trillium saponins can inhibit HT-29 and SW-620 proliferation possibly by affecting the expression and function of COX-2 and the expression of β-catenin.
