三裂叶野葛茎段和叶柄培养成再生植株 被引量:14

摘要 三裂叶野葛的叶柄和茎段外植体在含有 6 - BA1.0 mg/L 和 NAA0 .1m g/L 的 MS培养基上产生愈伤组织 ,愈伤诱导率分别为 10 0 %和 94% ,并进一步分化出芽 ,芽再生率分别为 10 0 %和 33.3%。而在添加 6 - BA3.0mg/L和 NAA 1.0 mg/L的 MS培养基中叶柄和茎段外植体的愈伤诱导率分别为 10 0 %和 6 4.3% ,芽再生率分别为 5 7.1%和 2 1.4%。芽长成嫩枝后转移至添加 0 .1m g/L NAA的 MS培养基后分化出根 ,形成小植株。再生植株的移栽成活率达 90 %。 The callus induction rates on MS medium containing 6-BA 1.0 mg/L + NAA 0.1 mg/L were 100% for petiole explant and 94% for stem explant. Callus could differentiate buds on the same medium, shoot regeneration rates were 100% and 33.3% respectively. While cultured in the medium supplemented with 6-BA 3.0 mg/L and NAA 1.0 mg/L, the callus induction rates were 100% and 64 3% respectively, and the shoot regeneration rates were 57.1% for petiole explant and 21.4% for stems. When the shoots were transferred onto MS medium containing 0.1 mg/L NAA, they differentiated into roots and formed regenerated plants. The survival rate was 90%.
