摘要
目的:阐明鸡胚胎发育过程中视顶盖层的形成以及神经元的迁移模式。方法:新鲜种蛋正常发育到第4.5天~第5天(E4.5-E5),将质粒pCAG-GFP通过活体原位电转技术转入视顶盖,继续培养到E8、E10和E12,取材、固定、包埋、切片后用PI复染核。结果:到E12鸡胚胎视顶盖从内向外最终形成6层结构,大部分神经元进行放射状迁移,部分神经元进行切向迁移,不同层的神经元有各自的形态特点。结论:鸡胚胎发育过程中不但存在由内向外的放射状迁移,还存在切向迁移模式。
Objective:To elucidate the migration mode of neurons and lamination in the chicken optic tectum during embryonic development.Method: Fresh eggs were cultured in normal condition at 4.5-5 days(E4.5-E5),pCAG-GFP plasmid were transfected into the optic tectum by electroporation in ovo.Tissues were harvested from E8 to E12 and fixed with 4% PFA,embeded with agarose,then cut with vibratome.The sections were counterstained with PI.Results:The optic tectum of chick embryo can be divided into six layers from the inside to outside,most of the neurons migrated radially,some neurons migrated tangentially,the neurons of different layer showed different morphological features.Conclusion:Neurons in optic tectum used different migration modes,radial migration and tangential migration.
