摘要 目的观察肿瘤坏死因子相关凋亡诱导配体(tumor necrosis factor related to apoptosis inducing ligand,TRAIL)联合白藜芦醇(resveratrol,Res)对人肝癌细胞凋亡的影响。方法流式细胞术检测细胞凋亡;CCK8法检测细胞增殖;Western blot检测细胞内Bax、Bcl-2蛋白的表达;分光光度法检测细胞内caspase3、caspase8的相对活性。结果 TRAIL联合Res作用Huh7细胞,联合用药组与对照组及单独用药组相比:肝癌细胞Huh7的凋亡率明显增加;促凋亡蛋白Bax表达明显增加,而凋亡抑制蛋白Bcl-2表达明显减少;caspase3、caspase8的相对活性明显升高。结论 TRAIL联合Res对诱导人肝癌Huh7细胞凋亡具有明显的协同效果,其机制可能与Bax表达增加、Bcl-2表达减少及caspase活性升高有关。 Human hepatoma carcinoma cell line (Huh7 cells) are resistant to tumor necrosis factor-related apoptosis-inducing ligand (TRAIL)-induced apoptosis, therefore, to investigated the effects of TRAIL in combination with resveratrol (Res) on Huh7 cell line, we examined the apoptosis of cells by flow cytometry, detected the proliferation by CCK8 assay, measured the expression of Bax and Bcl-2 by Western blot, and examined the relative activity of caspase3 and caspase8 by spectrophotometric method. Results showed that combined application of TRAIL and Res could promote Huh7 cell apoptosis, elevate Bax expression and the relative activity of caspase-3 and caspase-8, but suppress Bcl-2 expression, as compared with control group or single medication groups. From all the results we concluded that the TRAIL and Res have obvious synergistic effects in inducing apoptosis of hepatoma carcinoma cell line HuhT, and the mechanism may be associated with Bax, Bcl-2 and caspase activity.
