摘要
目的:建立琥乙红霉素静脉乳剂的含量测定方法。方法:采用Agilent Zorbax SB-C_(18)柱(250 mm×4,6 mm,5μm),流动相为乙腈-0.02 mol·L^(-1)醋酸铵缓冲盐溶液(65:35),检测波长215 nm,流速为1.0 ml·min^(-1),柱温25℃,进样量20μl。结果:琥乙红霉素在60~600μg·ml^(-1)的浓度范围内与峰面积呈良好的线性关系(r=0.999 1),平均回收率为97.92%,RSD为0.43%(n=9)。结论:该方法简便、快速、准确,可用于琥乙红霉素的含量测定及稳定性分析。
Objective:To establish an HPLC method for the determination of erythromycin ethylsuccinate intravenous emulsions. Method:An Agilent Zorbax SB-C_(18) column(250 mm×4.6 mm,5μm) was used.The mobile phase consisted of acetonitrile-0.02 mol·L(-1) ammonium acetate solution(65:35) and the detection wavelength was 215 nm.The flow rate was 1.0 ml·min(-1),the column temperature was 25℃and the injection volume was 20μl.Result:The linearity between the concentrations and the peak areas was obtained within the range of 60-600μg·ml(-1)(r =0.999 1).The average recovery was 97.92%(RSD = 0.43%,n = 9).Conclusion: The proposed method is simple,rapid and accurate,and suitable for the content and stability study of erythromycin ethylsuccinate intravenous emulsions.
