槐耳清膏对A549细胞增殖和自噬的影响 被引量:2

摘要 目的观察槐耳清膏对人肺腺癌细胞系A549细胞增殖和自噬的影响,探讨其作用机制。方法以A549细胞为研究对象,采用CCK-8法检测槐耳清膏对A549的增殖抑制率,用Westernblot方法检测自噬相关蛋白P62、LC3B表达的变化和mTOR信号通路分子表达的变化。结果0.5g/L及以上的槐耳清膏可明显抑制A549细胞的增殖,并且具有时间和浓度依赖性(P〈0.05)。用不同浓度(1、2、4、6、8g/L)的槐耳清膏作用A549细胞48h可以降低P62表达水平,增加LC3B的表达,降低mTOR信号通路关键分子的表达水平。联合使用自噬抑制剂3-MA可以加强槐耳清膏对A549细胞的增殖抑制率。结论槐耳清膏通过下调mTOR信号通路抑制A549细胞增殖,并且诱导A549发生自噬。联合使用槐耳和自噬抑制剂可能会更有效发挥槐耳抗肿瘤增殖的作用。 Objective To investigate the effects of Huaier on cell growth and autophagy in A549 cell line. Methods Cell growth was measured by CCK-8 assay kit, Western blot was used to test the autophagy effect and roTOR pathway. Results Huaier inhibited the proliferation of A549 in a time- and dose-dependent manner ( P ~ 0.05). Western blot analysis revealed cell autopbagy by LC3B H / I conversion and decreased P62. Western blot also indicated the downregulated mTOR signaling pathway induced by the increased concentration of Huaier. Combination treatment of autophagy inhibitor 3 MA and Huaier reinforced the inhibition effects. Conclusions Huaier inhibits the growth of A549 via downregulating roTOR pathway and induces autophagy. Combination therapy of Huaier and autophagy inhibitors are potentially more effective in inhibition of tumor cells than Huaier only.
