摘要
目的:观察海洛因依赖者外周血T淋巴细胞核仁形成区(NORs)酸性非组蛋白(Ag-NORs)表达及与解毒有关的胎盘型谷胱甘肽S-转移酶(CST-π)表达,分析海洛因依赖对T细胞代谢乃至功能的影响。方法:利用北京开隆公司提供的KL型免疫图象分析系统检测T细胞Ag-NORs;通过定量PCR技术分析病人外周血GST-π。表达。结果:72例海洛因依赖者外周血T淋巴细胞Ag-NORs检测平均值6.78%±0.50%(%±S),其中33例进行了GST-π表达分析,与正常人比较海洛因依赖者外周血GST-π表达明显增高,且Ag-NORs与GST-π表达呈明显负相关。结论:海洛因依赖者外周血T淋巴细胞Ag-NORs明显降低,表明其免疫细胞功能受到明显抑制。Ag-NORs降低的幅度明显小于肿瘤患者,说明海洛因依赖对T细胞的抑制可能在途径或机理上有别于肿瘤抑制因子对免疫细胞的抑制。GST-π表达增高,提示机体损伤修复机理加强。CST-π表达与Ag-NORs呈密切负相关,进一步证实海洛因依赖患者免疫功能损伤与修复对立调节的加强。
Objective: The expressions of T-cell nucleolar organizer regions(NORs) associated proteins(Ag-NORs) and blood Glutathione s-transferase-pi(GST-π) in heroin dependent individuals were analyzed by a KL Imaging System and a RT-PCR method. The results were compared with those of the control groups and malignant patients. Methods: Ag-NoRs of 72 cases of heroin dependent individuals were analyzed with a KL Imaging System, while the expressions of CST-π in 33 cases of heroin dependence were measured by quantitative PCR. Results: The AgNORs in heroin dependent individuals were 6.78% ±0.50% (x% ±s ) in average, The expression rate and level of GST-π were all significantly higher in heroin dependence than in control group, but no relationship between the GST-π mRNA and such as sex, dependent time and the type of drug taking was found. In addition, the reverse correlation between Ag-NORs and CST-π was found. Conclusion: The Ag-NORs in heroin dependent individuals were significantly lower than in normal adults, but still higher than in patients with malignant tumors. The results show that heroin dependence might induce immune damage, but the mechanism is diferent from which in patients with malignancies. The elevated expression of GST-π in blood cells demonstrated the elevation of the function of damage-repairmen in heroin dependent ivdividuals.
