可降解胸骨固定材料聚丁二酸丁二醇酯对THP-1细胞体外生物学特性的影响

摘要 目的:研究生物可降解胸骨固定材料聚丁二酸丁二醇酯(PBS)对THP-1细胞体外生物学特性的影响。方法:EdU和Hoechst 33342染色法分别研究PBS浸提液共培养后THP-1细胞的增殖和凋亡;RT-qPCR检测细胞凋亡相关基因Bax/Bcl-2和Smac/IAP-1的表达变化;染色体畸变试验评价其基因毒性。结果:PBS浸提液对THP-1细胞的体外增殖率影响较小,不存在细胞增殖半数抑制剂量;浸提液共培养后的细胞出现部分凋亡,基因表达改变以Bax/Bcl-2显著(P<0.05),Smac/IAP-1变化不明显(P>0.05);最大浓度浸提液共培养后的细胞染色体畸变率在正常范围。结论:PBS对THP-1细胞的体外生物学特性影响较小,对该细胞显示良好的生物相容性。 Objective To evaluate the effects of PBS on the biological characteristics of THP-1 cells in vitro.Methods The EdU and Hoechst 33342 staining were used to assess the proliferation and apoptosis of THP-1 cells co-cultured with the extracts of the materials respectively.The expression of Bax /Bcl-2 and Smac /IAP-1 related to apoptosis were evaluated by RT-qPCR.The genotoxicity of extract was evaluated by chromosomal aberration test.Results The extract of PBS showed little affection on the proliferation of THP-1 cells after seventy-two hours' incubation and 50% concentration of inhibition was not visible.The expression of Bax /Bcl-2 changed significantly(P〈0.05),however,the variations of Smac /IAP-1 were tiny(P〉0.05).The results of chromosomal aberration tests were in normal range at the same time.Conclusions PBS do lightly affect on the biological characteristics of THP-1 cells and it can be a promising biodegradable polymer for sternal fixation because of excellent biocompatibility.
