摘要
本文应用基因工程技术构建了可溶性H 2Kb 分子与绿色荧光蛋白 (GFP)融合基因sKb GFP。在COS7细胞中表达后 ,在内质网区域呈现明显绿色荧光。RT PCR结果显示融合基因获得了正确表达 ,蛋白印迹分析表明细胞内产生了预期大小的融合蛋白。细胞培养上清经亲和层析柱纯化后 ,获得了预期大小的重组蛋白。
Chimeric gene sK b-GFP was constructed by fusing the genes coding soluble H-2K b and green fluorescent protein using genetic engineering technology After transfection into COS7 cells,clear green fluorescence could be observed in the endoplasmic reticulum The result of RT-PCR analysis showed the correct expression of chimeric gene,and the Western blot demonstrated the existed protein of expected size Furthermore,the expected recombinant protein can be purified from culture supernatants by using the affinity column
