蓝舌病病毒在细胞内生长特性和在抗原快速纯化中的应用

摘要 为了解决原来蓝舌病病毒抗原提取中,步骤繁琐效率低的特点,研究了一种高效提取蓝舌病病毒抗原的方法。进行了蓝舌病病毒在细胞内生长特性的研究,采用酶染色等方法对不同接种量、不同培养时间的细胞内病毒含量进行测定,发现可以在感染细胞未产生病变前收获细胞,通过简单加工获得高浓度的病毒抗原。该方法快速高效,适用于蓝舌病的抗原生产,用于蓝舌病C-ELISA以及琼脂扩散实验中。该方法可推广用于其它病毒的纯化中。 For solving the problems of inefficient and time-cost in antigen purification of Blue-tongue virus antigen, a new method is reported here. At first,the concentrate of BTV in cells after inoculation was determined by IPX stain,then the best harvest time were decide when most f virus are in cells, not yet released in to culture medium. The harvested cells release high concentrate virus after frozen and thaw few times. The antigen got by this way has good quality and excellent to be use as rea- gent in C-ELISA and AGII3 test. The method could also he useful in other virus purification.
