摘要
为了合成新的、稳定的莱克多巴胺人工抗原,应用Mannich法将莱克多巴胺和载体蛋白BSA经一个亚甲基连接制备莱克多巴胺人工抗原,采用紫外光谱法、蛋白质电泳法、红外光谱法、双抗体夹心法和胶体金免疫层析法鉴定,证明莱克多巴胺人工合成抗原偶联成功、与抗莱克多巴胺抗体有特异性反应、具有抗原性。Mannich法合成的莱克多巴胺人工抗原未见报道,莱克多巴胺人工抗原用作包被抗原制成的莱克多巴胺快速检测试纸条对莱克多巴胺的检测灵敏度约为5 ng/mL,也可作为免疫源性抗原用于制备抗莱克多巴胺抗体。
To synthesis the novel and stable artificial racto- pamine antigen,Mannich reaction was used to conjugate rac- topamine and albumin from bovine serum (BSA) through a methylene to prepare the artificial ractopamine antigen. The conjugation was confirmed by UV Spectra, IR spectra and pro- tein electrophoresis. The binding reaction of the artificial rac- topamine antigen with anti-ractopamine antibody was con- firmed by double antibodies sandwich ELISA and gold immu- nochromatography assay (GICA). The results indicate that ractopamine-BSA can be successfully synthesized,and can be used as coating antigen in ractopamine rapid test strip with detection sensitivety of 5 ng/mL. Besides,the synthesized an- tigen could he used as immunogen to produce anti-ractopam- ine antibody.
