摘要
目的:探讨上皮性卵巢癌组织中雌激素受体β(ERβ)与磷酸化细胞外信号调控蛋白激酶1、2(p-ERK1/2)的表达及其与临床病理参数的关系。方法:选取2010年10月至2011年12月青岛大学医学院附属医院经病理证实的上皮性卵巢癌组织标本70例,卵巢良性肿瘤24例,正常卵巢组织24例。采用RT-PCR和免疫组织化学SP法检测上述组织中ERβ和p-ERK1/2的表达。结果:(1)卵巢癌组织中ERβmRNA相对表达水平(0.3764±0.9826)显著低于卵巢良性肿瘤(0.4900±0.3742)及正常卵巢组织(0.4980±0.0434)(P<0.05)。卵巢癌组织中ERβ表达与组织学分型、细胞学分级及淋巴转移有关(P<0.05);ERβ蛋白阳性表达率及其与临床病理特征关系与ERβmRNA一致;(2)ERK1/2 mRNA相对表达水平(0.6007±0.1554、0.6951±1.3694)显著高于卵巢良性肿瘤(0.3575±0.0479、0.5125±0.0411)及正常卵巢组织(0.3027±0.1024、0.5431±0.0811)(P<0.05);ERK1/2表达与细胞学分级、临床分期及腹水有关(P<0.05);p-ERK1/2蛋白阳性表达率及其与临床病理特征与p-ERK1/2 mRNA一致。ERβ蛋白与p-ERK1/2蛋白表达呈负相关(r=-0.282,P<0.05)。结论:ERβ低表达和p-ERK1/2高表达于卵巢癌组织,可能在卵巢癌的发生、发展中有重要作用。
Objective:To investigate the expression and the relationship between estrogen receptor β (ERβ)and phosphorylated extracellular signal-regulated protein kinase 1/2 (p-ERK1/2) in epithelial ovarian cancer. Methods :70 epithelial ovarian cancer specimen ,24 benign ovarian tumors and 24 normal ovarian tissues were obtained from epithelial ovarian cancer patients who were diagnosed in Affiliated Hospital of Qingdao Medical College from Oct. 2010 to Dec. 2011. Expression of ERβ, p-ERK1/2 mRNA and protein level were detected by RT-PCR and immunohistochemistry respectively. Results: ( 1 ) The expression of ERβ mRNA in patients with epithelial ovarian cancers (0. 3764±0. 9826) was significantly lower than that in benign ovarian tumors (0. 4900±0. 3742) and normal controls (0. 4980±0. 0434) ( P〈0.05 ). The expression of ERβ mRNA was closely related to pathologic grades and lymph node metastasis (P 〈0.05 ). The positive rates of ER[5 protein in ovarian carcinoma was well consistent with characteristics of ovarian carcinoma and expression level of its mRNA. (2)The expression of ERK1/ 2 mRNA in ovarian cancer (0. 6007±0. 1554,0. 6951± 1. 3694 ) was significantly higher than that in benign ovarian tumors (0. 3575 ±0. 0479,0. 5125 ±0. 0411 ) and normal controls (0. 3027 ±0. 1024,0. 5431 ±0.0811 ) (P〈0.05). The expression of ERK1/2 mRNA was closely related to pathologic grades, FIGO stages and presenee of ascites (P〈0.05). The positive rates of p-ERK1/2 protein were identical with characteristics of clinical diseases and expression level of its mRNA. Expression of ERβ was negatively correlated with expression of p-ERK1/2 in epi- thelial ovarian cancers (r=-0. 282 ,P〈0.05). Conclusion:The low expression of ERβ and over-expression p-ERK1/2 may play an important role in occurrence, development and progression of epithelial ovarian cancer.
