摘要
以酯交换量为指标,研究了醇溶液体系中蔗糖酯修饰猪胰脂酶的条件,并比较了修饰酶与未修饰酶的热稳定性.研究发现:30℃下40 min即可完成蔗糖酯对猪胰脂酶的修饰,但不同蔗糖酯修饰的最佳条件并不相同.S-370修饰的最佳条件是添加量5%,醇水比1∶3;S-570修饰的最佳条件是添加量5%,醇水比1∶2;S-770修饰的最优条件是添加量15%,醇水比1∶3;S-970修饰的最优条件是添加量20%,醇水比1∶3;S-1170修饰的最优条件是添加量15%,醇水比1∶3;S-1670修饰的最优条件是添加量5%,醇水比1∶3.热稳定性试验表明,6种蔗糖酯修饰的猪胰脂酶在40-80℃的催化反应活性都高于未修饰酶,在50-70℃修饰酶均有较高的催化活性.
Taking the transesterification rate as index, we studied the modification conditions of porcine pancreas lipase by sucrose esters in ethanol solution, and compared the thermal stability of the modified lipase and the original lipase. The results showed that the modification of porcine pancreas lipase could be completed at 30 ℃ in 40 minutes,but the modification conditions varied with the kind of the sucrose esters. The optimum modification conditions of 6 kinds of sucrose esters were as follows: S-370 amount 5% and ethanol-to-water ratio 1:3; S-570 amount 5% and ethanol-to-water ratio 1:2; S-770 amount 15% and ethanol-to-water ratio 1:3; S-970 amount 20% and ethanol-to-water ratio 1:3; S-1170 amount 15% and ethanol-to-water ratio 1:3; and S-1670 amount 5% and ethanol-to-water ratio 1:3. The thermal stability test showed that the catalytic activity of the lipases modified by the six kinds of sucrose esters was higher than that of the non-modified lipase at 40 ℃ to 80 ℃, and the modified lipases showed better catalytic activity at 50 ℃ to 70 ℃.
