摘要
目的探讨间充质干细胞(mesenchymal stem cell,MSC)移植对视网膜缺血再灌注损伤(retinal ischemic-reperfusion injury,RIRI)后凋亡相关基因p53、caspase-2表达的影响。方法将52只健康SD大鼠随机分为正常组(4只)、模型组(24只)、MSC移植组(24只)。前房加压法建立大鼠RIRI模型。贴壁筛选法体外培养SD大鼠MSC,模型组于RIRI后1h予以玻璃体内注射PBS缓冲液,MSC移植组注射MSC。各组SD大鼠分别于RIRI后2h、6h、12h、24h、48h、72h处死,免疫组织化学染色检测各组SD大鼠RIRI后p53及caspase-2的表达情况。结果正常组各时间点未见p53及caspase阳性表达。RIRI后2h、6h、12h、24h、48h及72h模型组p53阳性细胞数量分别为(20.50±1.71)mm-2、(26.98±4.23)mm-2、(31.74±0.58)mm-2、(50.63±0.67)mm-2、(48.02±0.69)mm-2、(38.90±0.95)mm-2,MSC移植组分别为(14.48±0.47)mm-2、(17.23±0.64)mm-2、(21.64±0.58)mm-2、(28.69±1.12)mm-2、(18.73±1.21)mm-2、(17.10±0.98)mm-2;模型组caspase-2阳性细胞数量分别为(9.20±0.54)mm-2、(78.93±0.88)mm-2、(391.10±5.62)mm-2、(687.25±6.24)mm-2、(491.75±2.75)mm-2、(153.75±7.63)mm-2,MSC移植组分别为(4.23±0.36)mm-2、(58.03±5.48)mm-2、(282.25±56.82)mm-2、(588.75±9.07)mm-2、(389.75±8.18)mm-2、(121.75±3.59)mm-2;与各时间点模型组比较,MSC移植组p53及caspase-2的表达均明显减少,差异均有统计学意义(均为P<0.05)。结论 RIRI后行MSC玻璃体内注射,可抑制p53及caspase-2的表达,减少视网膜神经节细胞的凋亡,对RIRI有一定的保护作用。
Objective To explore the effects of mesenchymal stem cell (MSC) transplantation on the expressions of I)53 and caspase-2 after retinal ischemia reperfusion injury(RIRI). Methods A total of 52 healthy SD rats were randomly divided into three groups: normal group ( n = 4 ), model group ( n = 24 ), MSC transplantation group (n = 24). The model of retinal ischemia reperfusion injury was made by increasing the intraocular pressure. MSC were cultured by adherent method in vitro. MSC in model group were injected with PBS buffer in the vitreous cavity 1 hour after injury. And rats in MSC transplantation group were injected with MSC. SD rats in each group were killed respectively at 2 hours,6 hours, 12 hours,24 hours,48 hours and 72 hours after RIRI. Then the expressions of p53 and caspase-2 in each group were detected by immunohistochemistry assay at 2 hours ,6 hours, 12 hours,24 hours,48 hours and 72 hours after RIRI. Results There was no positive expression of p53 and caspase-2 in normal group. The number of p53 positive cells in model group at 2 hours,5 hours,12 hours,24 hours, 48 hours and 72 hours after RIRI were ( 20.50 ± 1.71 ) mm^-2, ( 26.98 ± 4.23 ) mm^-2, (31.74±0.58)mm^-2,(50.63 ±0. 67)mm^-2, (48.02±0.69)mm^-2 and (38.90±0.95) mm^-2 ,respectively; MSC transplantation group were ( 14. 48 ± 0. 47 )mm^-2, ( 17.23 ± 0. 64)mm^-2, (21.64 ± 0.58) mm^-2, (28.69 ± 1. 12)mm^-2, ( 18.73 ± 1.21 )mm^-2 and (17. 10±0.98)mm^-2.
