摘要
目的观察中药四妙勇安汤活性部位对载脂蛋白E基因缺陷(ApoE-/-)小鼠动脉粥样硬化斑块形成的影响。方法 60只8周龄ApoE-/-小鼠随机分为6组:四妙勇安汤活性部位高剂量组[1g/(kg.d)]、中剂量组[0.5g/(kg.d)]、低剂量组[0.25g/(kg.d)]及辛伐他汀组[15mg/(kg.d)]、罗格列酮组[10mg/(kg.d)]、模型组,饲以高脂饲料。16周后,观察主动脉弓动脉粥样斑块形成程度;测定斑块面积与管腔面积之比;测定血清总胆固醇(TC)、三酰甘油(TG)含量。结果与模型组比较,四妙勇安汤活性部位高剂量组、辛伐他汀组和罗格列酮组小鼠动脉粥样硬化斑块面积/管腔面积比值明显减小(P<0.05或P<0.01)。与模型组比较,四妙勇安汤活性部位高剂量组和辛伐他汀组TC水平显著降低;四妙勇安汤活性部位高、中剂量组和辛伐他汀组TG水平明显降低(P<0.05)。结论四妙勇安汤活性部位能够抑制动脉粥样硬化形成,推测该作用与其促进脂质代谢和抗炎的机制有关。
Objective To observe the effect of active ingredient of Simiao Yong'an decoction(SYDAI) on atherosclerosis in ApoEgene knockout mice. Methods Sixty ApoE knockout mice in 8 weeks old were randomized into 6 groups.. Model group,simvastatin group, rosiglitazone group, SYDAI high dose group,SYDAI medium dose group,SYDAI low dose group. The high fat diet was fed for 16 weeks,then the area of atherosclerosis plaque and lumen were measured. The levels of serum total cholesterol (TC) and triglycarides (TG) were detected. Results Compared with model group, the ratio of plaque to lumen(PA/LA) in SYDAI high dose group, simvastatin group,rosiglitazone group were lower(P〈0.05 or P〈0.01). The levels of serum TC were lower in SYDAI high dose group and simvastatin group. The levels of serum TG were lower in SYDAI high dose group,SYDAI medium dose group and simvastatin group. Conclusion SYDAI can inhibit the formation of atherosclerosis plaque,which maybe related to its ability to promote the lipid metabolism and anti - inflammatory.
