摘要
目的:制备温敏类弹性蛋白多肽(ELPs)自组装微粒,并考察其温敏特性及对模型药物多西他赛的包载效能。方法:将ELPs冻干粉重新分散于冷的NaCl磷酸盐缓冲液中,利用配备电热温控装置的紫外-可见分光光度计,在波长350nm处测定ELPs溶液的光密度,升降温速率为1℃/min,计算相变温度。向ELPs溶液中加入多西他赛50%乙醇水溶液,混合完全,相变温度下孵育30min即得载药微粒,保温离心测定包封率并考察体外释放情况。结果:相变温度下自组装形成的载药微粒外观圆整,粒径约为1μm,包封率可达15.12%。结论:ELPs具有温敏特性,且可实现对多西他赛的包载。
Objective: To prepare self-assembled elastin like polypeptides(ELPs) particles, and to evaluate their thermosensitive characteristics and the potential for package of the model drug docetaxel. Methods: Lyophilized powder of ELPs was resuspended in ice-cold phosphate buffer saline of NaCl. The optical density was measured at 350 nm during both heating and cooling process at a rate of 1 ℃/rain through a UV-Vis spectrophotometer equipped with a temperature controller to calculate the phase transition temperature(Tt). To prepare docetaxel loaded ELPs microparticles, the 50% ethanol solution of docetaxel was added to the ELPs solution. The mixed solution was incubated for 30 rain and centrifugated under the T, to investigate the encapasulation efficiency and release in vitro property. Results: The diameters of the produced particles with general spherical appearance were approximately 1 μm, and the entrapment efficiency of 15.12% was achieved. Conclusion: Thermal properties and package capacity for docetaxel of the ELPs were confirmed.
