摘要
以濒危野生果树天山樱桃(Cerasus tianschanica Pojark)茎段为试材,在研究不同灭菌剂种类及灭菌时间对外植体材料的灭菌效果后,研究比较了不同培养基种类和不同生长调节剂浓度组合对天山樱桃茎段启动培养的影响,以建立天山樱桃茎段的初代培养体系。结果表明:外植体材料灭菌以酒精20s+HgCl212min为好,MS培养基中腋芽的萌发率高于B5培养基,初代培养的配方为MS+1.5mg/L 6-BA+0.1mg/L IBA+30g/L蔗糖+6g/L琼脂粉为佳。
Taking the stem of Cerasus tianschanica Pojark which was endangered plant as material,the effects of different sterilization and different sterilization time on the explants of Cerasus tianschanica Pojark were studied,and the influenceof different basic medias and different growth regulator composite concentrations on axillary proliferation wereinvestigated in order to establish the initial culture system of Cerasus tianschanica Pojark.The results showed that the.best sterilization of the explants was Alcohol 20 s-b-HgCI212 min and the germinationrate of axillary bud in MS basicmedia was higher than B5 basic media. The initial culture system of Cerasus tianschanica Pojark was MS+1. 5 mg/L6-BA+0. 1 mg/L IBA+30 g/L Sucrose+6 g/L Agar powder.
