摘要
以一品红(Euphorbia pulcherrima)"天鹅绒"为试材,在基本培养基(1/2MS+IAA(1.2mg/L)+琼脂(7.2g/L)+蔗糖(30g/L))中添加不同浓度三氯化镧(LaCl3),移栽后置于3℃低温条件下进行胁迫,研究了不同浓度LaCl3对低温逆境下一品红组培苗生理生化指标影响及低温胁迫下LaCl3对组培苗抗寒性调控机制。结果表明:适当浓度的LaCl3能不同程度提高游离脯氨酸(Pro)、可溶性糖、可溶性蛋白含量及超氧化物歧化酶(SOD)、过氧化物酶(POD)活性,降低低温胁迫后丙二醛(MDA)含量。LaCl3能通过增强渗透调节能力和细胞清除活性氧能力,促进体内保护酶活性提高,降低膜脂过氧化程度,最终明显提高其低温抵御能力。LaCl3适宜的最佳使用浓度为0.5~10mg/L。
Taking Euphorbia pulcherrima as test materials, the regulation mechanism of LaCl3 which was frost resistance under the stress of low temperature were discussed, in the basic medium (1/2 MS+ IAA (1.2 mg/L) + agar (7. 9 g/L) + sucrose (30 g/L)) adding different concentrations of LaCl3 ,the physiological and biochemical index influence of tissue culture seedling of Euphorbia pulcherrima were studied in different concentrations of LaCl3 at low temperature of 3℃ after transplanting. The results showed that the proper concentration of different degree LaCl3 could improve free Pro, soluble sugar, soluble protein content and SOD, POD activity, and reduce MDA content. LaCl3 could increase the protective enzyme activity through the enhancement of osmotic adjustment ability and osmoregulation and scavenging active oxygen and reducing the membrane lipid peroxidation level of Euphorbia pulcherrima under low temperature stress,resulting in an increase in the cold resistance of tissue culture seedling of Euphorbia pulcherrima. Appropriate concentration of LaCl3 was 0. 5-10. 0 mg/L.
