谷氨酸钠致胰岛细胞损伤的作用机制探讨

摘要 目的:探讨不同浓度的味精(monosodiumsodiumglutamate,MSG)对大鼠血清叫氨基丁酸(γ—Aminobutyricacid,GABA)水平及胰岛细胞凋亡的影响。方法:40只雄性wistar大鼠随机分为空白对照组和MSG低、中、高剂量组..对照组给予普通大鼠饲料,实验组给予MSG低、中、高剂量灌胃,于实验第30周和33周颈动脉窦取血后解剖取胰腺,检测对照组和MSG组血清GABA水平及胰岛细胞凋亡率的变化情况。结果:与对照组相比,实验组血清GABA浓度随MSG剂量的升高而逐渐降低,但仅高剂量组具有统计学意义(P〈0.01),而MSG组胰岛细胞的凋亡率则随着MSG剂量的增加而增加,高剂量组具有统计学意义(P〈0.01)。结论:MSG可引起血清GABA浓度降低以及胰岛细胞的凋亡率增高,表明MSG可能是通过GABA引起大鼠胰岛细胞损伤的发生。 Objective : To explore the change of serum y-Aminobutyric acid (GABA) level and the rate of Islet cell apoptosis induced by monosodium sodium glutamate (MSG) of different does. Methods : 40 male wisteria mouse were randomly divided into four groups : the control group and MSG low does, middle dose and high dose group. The control group was feed in normal and the experimental groups were feed MSG solution which contained different MSG does. Results : Serum GABA levels in experimental groups were lower than control group when MSG does increased, but the decrease was significant only in the high does group (P〈0.01). And the rates of Islet cell apoptosis in experimental groups were higher than the control group as MSG does increased, and the increase was significant only in the high does group (P〈0.01). Conclusions : MSG could induce the reduction of serum GABA level and increase the rate of Islet cell apoptosis, MSG induce diabetes mellitus may have high correlation with the change of GABA.
