摘要
目的了解深圳西乡街道五年来引起感染性腹泻的副溶血弧菌血清型分布、毒力因子及分子流行病学特征,为暴发疫情早期预警预测提供基础数据。方法用传统血清凝集法做血清型鉴定、用荧光PCR法检测毒力基因,再采用脉冲场凝胶电泳(PFGE)对副溶血弧菌进行分子分型。并用BioNumerics软件对PFGE分型图谱进行聚类分析。结果从4 825份标本中,分离出目标致病菌1 077株,其中病原菌副溶血弧菌851株,占79.01%。851株中,563株副溶血弧菌为O3:K6型,占66.15%,其次是O4:K8型,占10.34%。93.4%(795/851)的菌株是tdh+tlh+trh-菌株。对60株O3:K6型副溶血弧菌进行PFGE图谱分析共得到20种不同的PFGE图谱类型,各菌株图谱条带差异小于3条。结论深圳西乡街道五年来副溶血弧菌感染以O3:K6型为主,93.4%的菌株是tdh+tlh+trh-菌株。各菌株图谱具有同源性。
Objective To investigate the distribution of the serotypes of Vibrio parahaemolyticus inducing infectious diarrhea in Xixiang street, Baoan District of Shenzhen in the recent five years and to explore its virulence factors and molecular epidemiological characteristics, so as to provide basis for the early warning and forecast of the epidemic outbreak. Methods The serotypes of the isolates of Vibrio parahaemolyticus were identified by traditional serum agglutination assay. The virulence genes were detected by fluorescence PCR. Molecular typing was performed by pulse field gel electrophoresis (PFGE), and cluster analysis was made on the PFGE maps by BioNumerics software. Results Totally 1,077 targeted pathogenic bacteria were detected from 4,825 specimens, among which Vibrio parahaemolyticus accounted for 79.01% (851/1,077). The predominant serotype of Vibrio parahaemolyticus in this area was 03 : K6 (66.15 % ), and serotype 04 : K8 came next ( 10.34 % ). 93.4 % (795/ 851 ) of the strains had the virulence gene pattern of tdh + tlh + trh -. PFGE analysis of 60 strains of 03 : K6 type Vibrio parahaernolyticus found 20 different maps, and the difference among them was confined to three bands. Conclusions 03 : K6 type is the predominant serotype of Vibrio parahaemolyticus in Xixiang Street in Shenzhen. 93.4 % of the strains carry tdh + tlh + trh -. The isolates have homology.
