摘要
目的观察参附注射液对大鼠急性胃粘膜病变过程中的胃粘膜保护作用及胃组织TNF-!mRNA与IL-1"mRNA表达的影响,探讨参附注射液是否通过抗炎机制发挥胃粘膜保护作用。方法 30只成年Wistar大鼠随机分成3组,正常组(A组,n=10),应激组(B组,n=10),参附注射液预处理组(C组,n=10),在(20±1)℃水温下,B组、C组用浸水束缚应激法(WRIS)复制急性胃粘膜病变模型。6 h后处死取胃组织标本,10×解剖显微镜下观察各组胃粘膜损伤指数(GI),采用RT-PCR技术测定各组胃组织中TNF-!mRNA与IL-1"mRNA表达变化。结果(1)与A组比较,B组大鼠经WRIS 6h后胃粘膜损伤严重,GI明显升高(P<0.01);与B组比较,C组能显著减轻WRIS后胃粘膜损伤,降低GI(P<0.05)。(2)与A组比较,B组TNF-!mRNA与IL-1"mRNA表达显著上调(P<0.05);与B组比较,C组TNF-!mRNA与IL-1"mRNA表达显著下调。(3)GI的变化与TNF-!mRNA和IL-1"mRNA的变化呈正相关(r1=0.981,r2=0.992,P<0.05)。结论参附注射液预处理能明显下调大鼠急性胃粘膜病变过程中胃组织TNF-!m RNA与IL-1"mRNA的表达,阻抑应激后胃粘膜炎性反应可能是参附注射液发挥胃粘膜保护作用的重要机制之一。
Objective To investigate the gastroprotection of senfu injection(SFI) preconditioning and the effects on the expressions of TNF-oLm RNA and IL-113m RNA of gastric mucosal tis- sues in rats with acute gastric mucosal lesion (AGML) induced by water immersion and re- straint stress (WRIS) at 20± 1℃.Methods Thirty Wistar rats were randomly divided into 3 groups, including normal group (Group A,n= 10) ,WRIS stress group ( Group B,n= 10) ,SFI preconditioning and WRIS stress group ( Group C,n= 10). Gsatric mucosal lesion model was induced by water immersion and restraint stress (WRIS) at 20±1℃. After 6 h of WRIS, gastric tissues were excised and gastric lesion index(GI)was observed with the 10xanatomical microscope.The expressions of TNF-αmRNA and IL-1βm RNA of gastric mucosal tissues were de- tected by RT-PCR technology. Results WRIS for 6 h induced the severe lesion of rat gastric mucosa and significantly increased GI (P〈0.01) in addition to the rise of the expressions of TNF-αm RNA and IL-1βm RNA of rat gastric mucosa (P〈0.01). SFI preconditioning effectively ameliorated acute gastric mucosal lesion, lleviated GI and inhibited the higher expressions of TNF-αm RNA and IL-1βm RNA of rat gastric mucosal tissues in rats with acute gastric muco- sal lesion induced by WRIS, but did not completely avoid the gastric mucosal lesion after WRIS for 6 h. The changes of the expressions of TNF-αm RNA and IL-1βm RNA was positively cor- related with G I( = 0.892, r2 = 0.794, both P〈0.01 ). Conclusion SFI preconditioning can effectively ameliorate acute gastric mucosal lesion and alleviate GI, which decreased the mRNA expres- sion of proinflammatary factors in the process of AGML and protected the gastric mucosal cell probably by anti-inflammatory mechanism.
