摘要
以醋酸纤维素膜为基膜,采用环氧氯丙烷活化法活化基膜并在碱性条件下偶联磺胺二甲基嘧啶亲和配基,制备磺胺二甲基嘧啶亲和膜,对活化条件进行了优化并对其性能进行了表征。结果表明:膜上环氧基和配基含量分别为158.6μmol/g和114.6μmol/g,将其应用于腹水中分离纯化IgG,实验显示IgG在膜上的吸附平衡符合Langmuir方程,最大IgG吸附量为5.7 mg/g,一步分离IgG的纯度大于95%。
Using sulfamethazine as ligand,the sulfamethazine-affinity membrane was prepared and its application in purification of IgG was studied. Acetate membranes were activated with epichlorohydrin and coupled to sulfamethazine and, the density of epoxy groups and sulfamethazine in the membrance was 158.6 μmol/g and 114.6 μmoL/g, respectively. The obtained sulfamethazine-- affinity membranes were used to purify IgG from the ascites. The adsorption mechanism of IgG and the maximum IgG-binding capacity of the sulfamethazine-affinity membranes were investigated. The results suggested that the adsorption isotherm fitted the Langmuir model well and the membrance maximum adsorption capacity was 5.7 mg/g IgG/g (wet). Affinity purification process obtained a purity of above 95 %.
