摘要
目的探讨短期胰岛素治疗后糖尿病(DM)大鼠内脏脂肪组织内脏脂肪素(visfatin)、抵抗素(re-sistin)、脂联素(adiponectin)mRNA表达的变化及意义。方法雄性SD大鼠予小剂量STZ+高糖高脂饲料喂养制备为糖尿病大鼠,予胰岛素分别控制血糖在〈10 mmol/L(糖尿病血糖严格控制组)、10~14 mmol/L(糖尿病血糖非严格控制组)和〉16.7 mmol/L(糖尿病血糖未控制组)3个不同水平,同时设空白对照组。分别检测四组大鼠内脏脂肪组织visfatin、resistin、adiponectin mRNA的表达;并观察其游离脂肪酸(FFA)、甘油三酯(TG)、总胆固醇(TC)水平的变化。结果与血糖未控制组比较,血糖严格控制组FFA、TG均显著降低(P〈0.05),而TC显著增高(P〈0.05)。糖尿病血糖未控制组大鼠内脏脂肪组织visfatin、resistinm RNA表达均较空白对照组显著增高(P值均〈0.05),adiponectinm RNA表达显著降低(P〈0.05)。与糖尿病血糖未控制组比较,糖尿病血糖未严格控制组及严格控制组大鼠随着血糖的明显降低,visfatin mRNA表达显著下降(P〈0.05),resistin的表达逐渐下降,adiponectin的表达水平有所上升。大鼠内脏脂肪visfatin与resistin呈显著正相关(r值0.689,P〈0.05)。结论 visfatin、resistin及adiponectin可能在糖尿病大鼠糖脂代谢中发挥作用。
【Objective】To explore the mRNA expression changes and significant of visfatin,resistin and adiponectin in diabetic rats adipose tissue with short-term insulin treatment.【Methods】The DM rats were divided into 3 groups(the strict control group,the non-strict control group and the uncontrolled group) according to the target levels of blood glucose which were controlled by protamine zinc insulin,the visfatin,resistin and adiponectin mRNA expressions in visceral adipose in different blood glucose of diabetic rats were examined.The changes of serum visfatin,FFA,TG,TC levels of different blood glucose diabetic rats were observed.【Results】The levels of serum FFA,TG were decreased significantly in the group of strict control blood glucose compared with the group of uncontrolled blood glucose(P 0.05),yet the levels of TC increased significantly(P 0.05).The expressions of visfatin,resistin in diabetic rats adipose tissue in the group of uncontrolled blood glucose were significant increased compared with normal control group,the expression of adiponectin was significant decreased(P 0.05,respectively).The expression of visfatin was significant decreased(P 0.05),resistin was decreased;adiponectin was increased following with the decreasing of blood glucose.Visfatin was positive correlation with resistin in diabetic rat adipose tissue(r =0.689,P 0.05).【Conclusion】Visfatin,resistin and adiponectin maybe play roles in the metabolism of glycolipids in diabetic rats.
