阿托伐他汀钙对过氧化氢致内皮细胞损伤的保护作用

摘要 目的研究阿托伐他汀钙对过氧化氢(H2O2)致人微血管内皮细胞HMEC损伤的保护作用。方法采用H2O2损伤HMEC,研究阿托伐他汀钙对H2O2损伤的保护作用。用倒置相差显微镜观察细胞形态,采用酶活性测定法检测细胞培养上清中的乳酸脱氢酶(LDH),用ELISA法检测培养上清中ICAM-1及P-selectin的含量,MTT法检测细胞活力。结果 H2O2刺激HMEC后,细胞形态明显改变,细胞收缩,间隙变大;LDH释放明显增加(P<0.01);细胞活力明显降低(P<0.01)。用阿托伐他汀钙预处理后,细胞形态改变明显减轻,LDH释放降低(P<0.01),细胞活力增加(P<0.05)。结论阿托伐他汀钙对H2O2所致的内皮细胞损伤有保护作用。 Objective To investigate the protective effects of atorvastatin calcium against injury of hu- man microvascular endothelial cells induced by hydrogen peroxide. Methods Human microvascular endo- thelial ceils were treated with hydrogen peroxide and atorvastatin calcium. The morphological changes of the cells were observed with an inverted phase contrast microscope. LDH leakage was determined by en- zyme assay kit, and the content of P - selectin and ICAM - 1 in supernatants was assayed by ELISA kits. Finally, the cell viability was measured by MTI assay. Results After exposure to H2 02, endothelial cells obviously deformed and shrinked. Moreover, the release of LDH was also significantly increased. Con- versely, cellular viability was decreased.
