摘要
以“晖雨露”水蜜桃为试验材料,建立了一种适用于桃果实总蛋白质的双向电泳体系。最终试验确定采用酚抽法提取桃果实蛋白质,采用17cm,pH4-7的线性IPG胶条,800μg蛋白质上样,并用考马斯亮蓝G-250胶体考染,可以得到背景清晰、分辨率高的双向电泳图谱。对影响双向电泳图谱质量的因素进行了讨论分析。
An effective two-dimensional electrophoresis system was developed to determine the total protein from "huiyulu" peach fruit in this study. The optimum conditions for total protein detection were listed as follows: phenol extraction,800μg protein sample, 17 cm IPG strip with pH 4-7,and finally detecting proteins with modified colloidal coomassie brilliant blue G-250 staining. With the optimum conditions,a high quality 2-DE map with low background was achieved.
