摘要
丝氨酸蛋白酶抑制剂(Serpin)在昆虫的先天性免疫及生长发育中发挥重要作用。为研究家蚕丝氨酸蛋白酶抑制剂6(Bmserpin-6)在家蚕表皮黑化反应、蜕皮变态和先天性免疫等生理过程中的功能作用,采用实时荧光定量PCR方法,检测Bm-serpin-6 mRNA在家蚕幼虫组织和蜕皮过程以及免疫刺激处理后的转录特征。结果表明:Bmserpin-6 mRNA在4龄幼虫的头部、表皮、中肠、丝腺和脂肪体中有转录,其中在表皮中高水平转录,但在眠中的转录水平下调;在5龄幼虫头部的转录水平随发育时间呈先升后降的趋势;脂多糖(LPS)刺激5龄第3天幼虫后,Bmserpin-6 mRNA在血液和脂肪体中的转录水平上升,其中在脂肪体中的转录水平自刺激后6 h达到最高值,在血液中的转录水平自刺激后9 h达到最高值。结果提示,Bmserpin-6可能与家蚕的表皮黑化反应及蜕皮有一定关系。
Serine protease inhibitors(Serpins) play important roles in innate immunity,growth and development of insects.In order to explore the roles of Bombyx mori serine protease inhibitor 6(Bmserpin-6) in epidermis melanization,molting and innate immunity of Bombyx mori,we employed real-time fluorescent quantitative PCR method to detect the transcriptional profile of Bmserpin-6 in larval tissues,during molting process and after immune stimulation.The results showed that there was transcription of Bmserpin-6 mRNA in head,epidermis,fat body,midgut and silk gland of the 4th instar silkworm larvae,among which the transcriptional level was high in epidermis though down-regulated during molting.The transcriptional level in head of the 5th instar silkworm larvae showed a first rise and then fall tendency.The transcriptional levels were increased in the hemolymph and fat body of day 3 larvae of the 5th instar induced by lipopolysaccharide(LPS).The highest transcriptional level was observed in fat body at 6 h after LPS injection,and the highest level was observed in hemolymph at 9 h after LPS injection.The result indicated that Bmserpin-6 has related physiological functions in epidermis melanization and molting of Bombyx mori.
