摘要
目的研究LY-294002对急性脊髓损伤大鼠Bcl-2表达的变化。方法选取104只成年健康SD大鼠,按Nystrom法建立大鼠脊髓(T8、T9)急性压迫损伤模型,按随机数字表法分为假手术组8只、损伤组48只、LY-294002组48只。免疫组化和Western印迹方法检测各组大鼠脊髓损伤部位Bcl-2的表达。结果免疫组化结果发现,假手术组大鼠脊髓有大量的Bcl-2阳性表达的神经细胞;而在脊髓损伤后2 h时,Bcl-2阳性细胞数已开始明显减少,在3 d时Bcl-2阳性表达细胞数最少,5 d时Bcl-2阳性表达细胞数开始增加。在经LY-294002处理后的各时间点大鼠脊髓损伤部位的Bcl-2阳性细胞明显高于相同时间点的脊髓损伤组大鼠。Western印迹方法检测结果与免疫组化方法一致。结论 LY-294002能够上调急性脊髓损伤大鼠Bcl-2蛋白的表达。
Objective To study the effect of LY294002 on the expression of Bcl-2 after acute spinal cord injury(SCI).Methods 104 SD healthy adult rats were chosen and randomly divided into sham(n=8),SCI(n=48) and LY-294002(n=48)groups.SCI was induced by Nystrom's way by the moderate compression at the level of T8 and T9 spinal cord.The expression of Bcl-2 was observed by immunochemistry and Western blot.Results There were plenty of Bcl-2 positive cells in sham group.The number of positive cells of Bcl-3 began to decrease obviously at 2 hours,was the lowest at 3 days,and began to increase at 5 days.Compared with SCI group,the number of positive cells of Bcl-2 at the same time point was significantly higher in injury sites of rats in LY-294002 group.The results of Western blot were coincidence with immunochemistry results.Conclusions LY-294002 could up-regulate the expression of Bcl-2 of rat after acute SCI.
