摘要
为全面了解兰州鲇(Silurus lanzhouensis)肌肉品质,建立了兰州鲇肌肉中肌苷酸含量的高效液相色谱测定方法。样品经5%pH 6.5~6.8的高氯酸溶液提取,C18反相色谱柱分离,254 nm波长处测定,外标法定量,测定了兰州鲇、鲇(Silurus asotus)、黄河鲤(Cyrinus carpio)3种鱼类肌肉中的肌苷酸含量。结果显示:该方法线性关系良好,相关系数r≥0.999;灵敏度高,检出限为2.5×10-4g/kg;加标回收率≥94.0%;方法精密度好,RSD≤2.5%。兰州鲇、鲇和黄河鲤的肌苷酸含量分别为1.18 g/kg、0.733 g/kg和0.504 g/kg。
For a comprehensive understanding of the meat quality of Silurus lanzhouensis, a method for the determination of inosine 5'-monophosphate(IMP) in S. lanzhouensis by high-performance liquid chromatography was establshed. IMP in Silurus lanzhouensis were extracted by 5% perchloric acid with the pH of 6. 5 -6. 8, separated by C18 reversed phase col- umn, and detected at wavelength of 254 nm by means of external standard method to detect IMP contents in three fishes of S. lanzhouensis, Silurus asotus and Cyrinus carpio. The results suggested: This HPLC method is simple and rapid to detect IMP contents of S. lanzhouensis, IMP was shown a good linear relation in the range of 0. 010 - 1.00 mg/mL( r≥0. 999 ), the limit of detection was 2. 5×10^-4g/kg, the recovery of the method was no less than 94.0% , RSD was smaller than 2. 5%. The method is accurate and sensitive, it can be used to determine the IMP in freshwater fish. The IMP quantity in freshwater fish of S. lanzhouensis, S. asotus and C. carpio were 1.18 g/kg, 0. 733 g/kg and O. 504 g/kg; respectively.
