摘要
背景与目的:实验证明MMP-9与IL-17可促进各种肿瘤细胞增殖及侵袭发展,但有关MMP-9与IL-l7是否共同参与了垂体腺瘤的发生、尤其是否与侵袭性有关,国内外尚无相关报道。本研究通过检测MMP-9和IL-l7在垂体腺瘤组织中的表达,探讨二者与垂体腺瘤发生发展的关系。方法:收集75例人垂体腺瘤组织,应用免疫组化和RT-PCR检测IL-17、MMP-9在肿瘤组织中的表达情况。结果:免疫组化结果显示,MMP-9在侵袭性垂体腺瘤中的强阳性及总阳性表达率分别为68.7%和91.7%,明显高于非侵袭性垂体腺瘤中的17.4%和57.7%(P<0.05);IL-17在侵袭性垂体腺瘤中的强阳性及总阳性表达率分别为41.5%和81.7%,明显高于非侵袭性垂体腺瘤中的17.7%和50.97%(P<0.05)。RT-PCR显示侵袭性垂体腺瘤组IL-17、MMP-9 mRNA表达水平明显高于非侵袭性垂体腺瘤组。结论:IL-17、MMP-9的高表达与垂体腺瘤的侵袭性密切相关,并且共同促进垂体腺瘤侵袭性生长,二者可作为垂体腺瘤侵袭性的有效指标。
BACKGROUND & OBJECTIVE: It has been shown that MMP-9 and IL-17 can promote tumor cell proliferation, development and invasion, hut it is not well known about whether MMP-9 and IL-17 participate in the development of pituitary tumors. This study was designed to investigate expression of MMP-9 and IL-17 in pituitary tumors and to explore the possible mechanisms of occurrence and development of pituitary tumors. METHOD: The expression of MMP-9 and IL-17 in pituitary tumors was detected by immunohistochemistry and RT- PCR. RESULT: The expression of MMP-9 and IL-17 in the invasive type of pituitary tumors were significantly higher than that in non-invasive pituitary tumors, there was a significant difference (P〈 0.05). CONCLUSIONS: The high expression of IL-17 and MMP-9 is associated with the development and invasion of pituitary tumors, and these might be used as markers for evaluating invasion potential.
