通窍化栓汤对脑缺血再灌注模型大鼠脑组织抗氧自由基的影响 被引量:6

Effects of Tongqiao Huashuan Decoction on Antioxidant Free Radical in Brain Tissues of Cerebral Ischemia Reperfusion Model Rats

摘要 目的研究通窍化栓汤对脑缺血再灌注模型大鼠脑保护的可能作用机制。方法将50只局灶性脑缺血再灌注模型大鼠随机分为模型组及通窍化栓汤组高、中、低剂量组和丹参注射液组,另设假手术组只进行麻醉和血管分离术,各组均10只。通窍化栓汤高、中、低剂量组分别按17.50、8.75、4.35g/(kg·d)灌胃通窍化栓汤,丹参注射液组给予丹参注射液5.25g/(kg·d)腹腔注射,模型组给予等量生理盐水腹腔注射,假手术组仅抓取,各组每日1次,共15天。测定各组大鼠脑组织丙二醛(MDA)及超氧化物歧化酶(SOD)含量。结果与模型组比较,其余各组大鼠脑组织MDA含量降低,SOD水平升高,差异有统计学意义(P<0.05或P<0.01);与通窍化栓汤高剂量组比较,通窍化栓汤中、低剂量组及丹参注射液组MDA水平升高,丹参注射液组SOD水平降低,差异均有统计学意义(P<0.05)。结论通窍化栓汤可能通过降低脑缺血再灌注模型大鼠脑组织MDA水平,升高SOD水平,从而减轻脑缺血损害。 Objective To study the protective effect and mechanisms of Tongqiao Huashuan Decoction (decoction for orificefree and thrombolysis) on cerebral ischemia reperfusion model rats. Methods Fifty model rats were randomized into model group, Tongqiao Huashuan Decoction high-dose, middle-dose and low-dose groups and salvia injection group, with 10 in each. Another 10 healthy SD rats were selected as sham operation group. The Tongqiao Huashuan Decoction high-dose, middle-dose and low-dose groups were gavaged with 17.50 g/(kg ·d), 8.75 g/(kg · d) and 4.35 g/(kg · d) Tongqiao Huashuan Decoction respectively. The salvia injection group was intraperitoneally injected with 5.25 g/(kg · d) salvia injection. The model group was intraperitoneally injected with normal saline. The rats in the sham operation group were caught only. Each group was ad- ministered once daily for 15 days. The contents of malondialdehyde (MDA) and superoxide dismutase (SOD) in brain tissues were detected. Results Comparing with the model group, the contents of MDA were significantly decreased but the contents of SOD were significantly increased in other groups (P〈0. 05 or P〈0.01). Comparing with the Tongqiao Huashuan Decoction high-dose group, the contents of MDA in the Tongqiao Huashuan Decoction middle-dose and low-dose groups and salvia injec- tion group were significantly increased but the content of SOD in the salvia injection group was significantly decreased (P~ 0.05). Conclusion Tongqiao Huashuan Decoction can reduce ischemic brain damage by decreasing the content of MDA and in- creasing the content of SOD in brain tissues of cerebral ischemia reperfusion model rats, especially with high dose therapy.
