摘要
目的:研究淫羊藿防治去卵巢大鼠骨质疏松症的作用及初步机制。方法:雌性7月龄SD大鼠分为正常组,模型组和淫羊藿高、低剂量组共4组(n=8),除正常组行假手术外,其余均手术彻底摘除卵巢,术后进行模型筛选,1周后依照正常组、模型组灌胃生理盐水,淫羊藿高、低剂量组灌胃淫羊藿提取液(剂量以生药量计为50,25 g.kg-1),连续给药90 d。给药45 d时检测血清钙(Ca2+),90 d时检测血清Ca2+、血清磷(P)、超氧化物歧化酶(SOD)、丙二醛(MDA)、碱性磷酸酶(ALP)活性、血清雌二醇(E2)和睾酮(T)含量,并分离胫骨进行病理切片观察。结果:与模型组血清Ca2+(3.01±0.33)mmol.L-1相比,给药45 d后低、高剂量淫羊藿组血清Ca2+均下降[(2.48±0.39),(2.86±0.34)mmol.L-1,(P<0.05,P<0.01)],90 d后高剂量淫羊藿组血清Ca2+下降更明显(1.76±0.53)mmol.L-1,(P<0.01)。与模型组相比,淫羊藿能降低血清MDA(8.4±1.3)mmol.L-1(P<0.01),提高血清P(4.61±0.82)mmol.L-1(P<0.05),SOD活性(110±13)U.L-1(P<0.05),碱性磷酸酶活力(4.4±0.89)U.L-1(P<0.05),雌二醇水平(25.66±9.38)pg.L-1(P<0.05)和睾酮水平(1.95±1.21)ng.L-1(P<0.05)。胫骨病理切片显示高剂量淫羊藿能预防骨小梁断裂,Ⅰ级和Ⅱ级病变率占87.5%,无Ⅳ级病变。结论:淫羊藿明显提高性激素水平,清除活性氧,可能是其防治骨质疏松症的主要机制。
Objective: To study the effect of Epimedii Folium on ovariectomy-induced osteoporosis in rats. Method: The female SD rats were divided into sham operated group (Sham), ovariectomized group (Model) , high concentration Epimedii Folium with ovariectomized group (H-EF) and low concentration Epimedii Folium with ovariectomized group (L-EF). The serum Ca2+ were examined after administration for 45 days and 90 days. And the serum P, superoxide dismutase (SOD), malonaldehyde (MDA), estradiol (E2), testosterone (T) and alkaline phosphatase (ALP) were examined after 90 days ofadministration. The tibial slice was also analysized. Result: Compared with the model group, the serum Ca2+ was decreased [ (2.48±0.39) mmol·L-1 (P 〈 0.05), (2.86±0.34) mmol .L-1 (p 〈 0.01 ) ] by epimedium after 45 days of administration, and the serum MDA (8.4+1.3) mmol·L-1 (P〈0.01) for90 days. Butthe serum ALP (4.4±0.89) U·L-1(P〈0.05), P (4.61±0.82) mmol-L-1 (P〈0.05) E2 (25.66±9.38) pg.L-1 (P〈0.05), T (1.95±1.21) ng.L-1 (P〈0. 05) and SOD (110±13) U·L-1 (P〈0.05) were all increased by hi Epimedii Folium. Conclusion: It may be one of the mechanisms on osteoporosis that Epimedii estrogen and decrease ROS.
