摘要
目的:探讨左归丸对5/6肾大部切除模型并肾性骨病大鼠甲状旁腺功能亢进的影响。方法:SPF级Wistar大鼠进行5/6肾大部切除并给予高磷饮食诱导肾性骨病模型。造模大鼠分为模型组、左归丸组、骨化三醇组,并设正常组、假手术组。药物干预12周处死大鼠,检测血清全段甲状旁腺激素(iPTH);双能X线测量大鼠股骨骨密度(BMD);分别采用Real-time PCR、western blotting检测甲状旁腺PTH mRNA与蛋白的表达;免疫组化的方法检测甲状旁腺PCNA的表达。结果:12周时,模型组大鼠出现了血iPTH升高,甲状旁腺增生,骨密度下降。左归丸组大鼠iPTH水平与模型组比较(P<0.01)明显下降,左归丸组大鼠骨密度与模型组比较明显改善(P<0.01),明显抑制甲状旁腺增生。结论:左归丸能降低5/6肾大部切除模型并肾性骨病大鼠甲状旁腺素合成以及血iPTH水平,抑制甲状旁腺增生,改善肾性骨病。
Objective: To investigate of the beneficial effects of Zuoguipills on secondary hyperparathyroidism in 5/6 nephrectomy-induced renal osteodystrophy(ROD)rats.Methods: 24 rats were 5/6 nephrectomized and fed with high dietary phosphorus,the other 8 rats were dealt with pseudo-operation.The nephrectomized rats were divided into model group,Zuoguipill group,and Calcitriol group(n=8 in each group).The other 8 rats were normal group.The rats were sacrificed at the end of 12 weeks' treatment.The blood iPTH was measured;the expression of PTH mRNA and protein in parathyroid gland were measured by real-time PCR and western blotting,respectively;the femur bone mineral density(BMD) was measured by dual energy X-ray.Result:In the model group,blood iPTH levels were significantly increased,and BMD was decreased.Blood levels of iPTH was significantly lower and BMD was significantly higher in Zuoguipill group than that in model group(P0.01).The expression of PTH mRNA and protein were significantly lower in Zuoguipill group than that in model group(P0.01).Compared with model group,group treated with Zuoguipill ameliorated parathyroid hyperplasia and improved BMD(P0.01).Conclusion: Zuoguipills can ameliorate hyperparathyroidism and renal osteodystrophy.The treatment efficacy perhaps has the relation with ameliorating parathyroid hyperplasia and decreasing iPTH level.
