摘要
目的 :制备抗粘放菌 5 5 19Ⅰ型菌毛单克隆抗体。方法 :采用BALB/C小鼠免疫后取脾细胞与SP2 /0小鼠骨髓瘤细胞杂交融合 ,制备单克隆抗体 ,并以ELISA法和双向琼脂扩散法进行鉴定。结果 :共得到 6株杂交瘤细胞 ,抗体效价达 1:10~ 10 0万 ,类型为IgG ,亚类为IgG1。结论 :经BALB/C小鼠免疫可得到高纯度的抗粘放菌Ⅰ型菌毛单克隆抗体 ,而且效价高 ,细胞株稳定分泌抗体。
Objective: Preparation of the monoclonal antibody against type I fimbriae of A. Viscosus 5519. Method: BALBA/C mice were administered antigen of fimbriae. Then the spleens of mice were removed and hybridized with cells of SP2/0, monoclonal antibody were prepared. The monoclonal antibody was identificated with ELISA and double agar gel diffusion. Results: Six hybridomas were obtained. Titre of ascites fluid reached 100,000~1,000,000. Type of antibody was IgG, subtype was IgG1. Conclusion: An monclonal antibody against type I fimbriae of A. Viscosus can be obtained by BALB/C immunity. Titre of antibody not only is high, but antibody secreting of cells is steady.
