摘要
根据主流酶法脱墨机制,经富集、初筛和多轮复筛,成功的从造纸厂采集的土样中,分离得到了5株具有良好脱墨能力的菌株。采用液体发酵的方法,从这5株微生物中筛选得到了一株脱墨能力最好的菌株M03-#3-001。形态学观察以及ITS序列分子鉴定结果表明,该菌属于烟曲霉属。M03-#3-001菌株在100L发酵罐上的研究结果显示,发酵时间为84h,脱墨效果最佳,白度(ISO)增加值可达到3.42,此时发酵液CMC酶活力为855.23U/ml,α-淀粉酶活力为5683.37U/ml;对该菌株的脱墨条件进行研究,结果显示脱墨反应的适宜时间为40min,适宜pH为8.0,适宜温度为30℃。
According to the main mechanism of enzymatic-deinking of waste paper, five deinking strains were isolated from the soil sample provided by Nanping Paper Co., Ltd. Using the method of liquid fermentation, one strain(M03-#3-001) with best deinking ability was screened from the five strains of microorganisms. Morphological observation and ITS sequence identification results show that M03-#3-001 belongs to Aspergillus fumigatus. The results of M03-#3-001 strains in 100 liter fermentation tank show that the effect of deinking is optimal on the condition of fermentation time of 84 h, the brightness (ISO) increment reaches 3.42, then the activity of CMC-ase is 855.23U/ml, the activity of a-amylase is 5683.37U/ml. Deinking conditions of M03-#3-001 was studied, the results show that the best deinking reaction time of 40min, the optimum pH 8.0, the optimum temperature 30℃.
