摘要
目的:建立1种液相色谱-质谱联用法测定大枣中三萜酸的含量。方法:采用聚合物键合的ODS色谱柱Ulti-mate XB-PAH色谱柱(4.6 mm×250 mm,5μm)进行分离,以乙腈-水(90∶10)为流动相,电喷雾质谱负离子选择离子模式检测,对不同产地大枣中的白桦脂酸、齐墩果酸和熊果酸3种三萜酸成分进行了测定和比较。结果:所建立的检测方法灵敏度高(50.8~113.9 pg),分离度好(R≥1.7),线性关系良好(r≥0.999 0),重复性好(RSD≤3.5%)。结论:聚合物键合的ODS固定相可以对三萜酸同分异构体进行高效分离,并且HPLC-MS对大枣中的三萜酸含量分析,专属性和灵敏度显著提高,可满足实际检测需要。
A simple and sensitive method has been developed to simultaneously determine betunilic acid, oleanolie acid and ursolic acid in the fruits of Ziziphusjujuba from different regions by HPLC-MS. This HPLC assay was performed on PAH polymeric Cls bonded stationary phase column with mobile phase contained acetonitrile-water (90: 10) and with negative ESI detection mode. The developed approach was characterized by short time consumption for chromatographic separation, high sensitivity and good reliability so as to meet the requirements for rapid analysis of large-batch fruits of Z. jujuba from different habitats.
